Movement of birds that eat berries and plastid DNA diversity of the mistletoe (Viscum) in Central Europe

Abstract:

Jmelí bílé (Viscum album) patří mezi poloparazitické rostliny vyskytující se na území Evropy. Zatímco v některých oblastech jej nenalezneme, v jiných má expanzivní, až vůči hostiteli letální chování. Cílem práce bylo přispět různými pohledy k porozumění existence těchto kontrastních vzorů. Vztahy v rámci V. album byly studovány prostřednictvím haplotypových sítí; z plastidové DNA regionu trn bylo analyzováno 92 vzorků a z jaderného regionu ITS 40 vzorků jak z vlastních sběrů, tak sekvencí z databáze GenBank. Haplotypová síť regionu trn odhalila dominantní panevropský haplotyp V. album album s hvězdicovou strukturou okolních haplotypů naznačující recentní expanzi poddruhu. Poddruh V. album austriacum vytvářel dvě oddělené skupiny; jednu s kontinentální distribucí a druhou s haplotypy endemizujících linií napříč Středozemím. Taxony V. album abietis, V. album cruciatum a V. creticum naznačovaly retikulátní historii. V jaderném markeru ITS tvořili jedinci poddruhu V. album album dva dominantní haplotypy bez výrazné geografické strukturace nejen ve střední Evropě, ale také napříč kontinentem včetně střední Evropy, ač jedna linie měla svého zástupce v západní Evropě a druhá naopak ve východní. Taxony V. album abietis, V. album austriacum vytvářely samostatné skupiny, podobně jako jedinec považovaný za V. album album z Japonska, vzdálený od evropské skupiny V. album album 20 mutačními kroky. Vzor distribuce haplotypů mohla komplikovat historická introdukce jmelí člověkem na nové lokality a homogenizace markeru ITS procesem "concerted evolution". Vysoká invazivita ve vztahu k hostiteli je doménou poddruhu V. album album. Role bobuložravých ptáků na šíření jmelí má minimálně lokální charakter a pro další studium vztahů je nutno k areálovým mapám získat také data o hustotě výskytu jak ptáků, tak taxonů rodu Viscum.

Abstract:

European mistletoe (Viscum album) belongs to a group of hemiparastic plants found mainly in Europe. In some areas it cannot be found, whereas in other areas it is abundant, to the point of being lethal to its host. The goal of this thesis was to analyze and understand the contrasting behaviour exhibited by the plant. The relationships of V. album were studied via haplotype networks; 92 samples were analyzed from the plastid trn DNA region and 40 samples from the ITS nuclear region. These samples came from our own collection and GenBank sequences. The haplotype network of the trn region revealed a dominant pan-European haplotype (V. album album) with a star-like structure of the surrounding haplotypes indicating the recent subspecies expansion. The subspecies V. album austriacum created two sub groups; one which had a continential distribution and another with haplotypes of endemic lines across the Mediterranean. The following taxons of V. album abietis, V. album cruciatum, V. creticum exhibited a reticulated history. The ITS nuclear marker revealed two subspecies of V. album album in which two dominant haplotypes existed without distinctive geographic structures across Europe, although one genetic lineage had its representatives in Western Europe and the other in the East. The taxons V. album abietis and V. album austriacum created individual groups similar to V. album album from Japan, which is separated from the european group of V. album album by 20 mutations. The haplotype distribution pattern could be complicated by the historical anthropogenic introduction of mistletoe to new sites and homogenization of the ITS marker by the "concerted evolution" process. The high invasiveness in relation to the host is caused by the subspecies V. album album. Berry-eating birds affect mistletoe propagation in the local area. For the further study is necessary to get enough data about the density of birds and taxons of Viscum.
