Detection of binding of the splicing regultory proteins on the pre-mRNA and the determination of the differences in this binding upon selected human pathological mutations

Abstract:

The pre-mRNA splicing is a fundamental step in the expression of genes by which introns are removed from the pre-mRNA and exons are joined to form a mature transcript. The importance of an accurate splicing is illustrated by the fact that at least 15 % up to 50 % of splicing mutations cause human genetic diseases. In this thesis I deal with c.1092T›A mutation in the gene encoding iduronate-2-sulfatase (IDS) that causes the X-linked recessive lysosomal disease, mucopolysaccharidosis II (MPS II). The c.1092T›A leads to the use of multiple cryptic splice sites and produces multiple aberrant transcripts. In order to understand the molecular mechanism which results in aberrant splicing combinations of experimental analyses are used. Their results show that the most probably mechanism of the splicing deregulation with observed mutation is the creation of new splicing silencers and demonstrate the complexity of the splicing regulation. In addition, the results emphasised the importance of the study pre-mRNA splicing and it is necessary to perform more studies focusing on splicing regulatory elements in order to get better understanding of their function.

Abstract:

Sestřih pre-mRNA je základní krok genové exprese, kterým dochází k odstranění intronů z pre-mRNA a k následnému spojení exonů za tvorby zralého transkriptu. Důležitost přesného sestřihu je vyzdvižená faktem, že 15 až 50 % sestřihových mutací způsobuje lidská dědičná onemocnění. V této práci se zabývám mutací c.1092T›A v genu kódujícím enzym iduronát-2-sulfatázu (IDS), která způsobuje X-vázanou recesívní lyzozomálnu chorobu, mukopolysacharidózu II (MPS II). Mutace c.1092T›A vede k využití mnoha kryptických míst sestřihu a vytváří tak několik aberentních transkriptů. Pro pochopení molekulárního mechanismu, kterým dochází k aberentnému sestřihu, byly použity kombinace experimentálních analýz. Jejich výsledky ukázaly, že nejpravděpodobnějším mechanizmem deregulace sestřihu sledovanou mutací je vznik nového zeslabovače sestřihu, a poukázaly na komplexitu sestřihové regulace. Mimo to výsledky zdůraznily důležitost studia sestřihu pre mRNA a nutnost provádět více studií sestřihových regulačních elementů pro lepší pochopení jejich funkce
