Autophagy Induction and Detection \kur{in vitro}

Abstract:

Tato bakalářská práce popisuje proces autofagie včetně mechanismů její indukce a detekce. První část je věnována proteinům a proteinovým komplexům, které jsou pro tento proces nezbytné. V hlavní části práce jsou uvedeny způsoby indukce autofagie se zaměřením na \kur{in vitro} systémy. Shrnuty jsou jak chemické, tak biologické způsoby indukce zahrnující například látky jako je lithium, trehalóza nebo metformin. V další části práce je pak charakterizována obrazová cytometrie, transmisní elektronová mikroskopie a vybrané typy imunoanalýzy, jakožto metody detekce autofagie v \kur{in vitro} systémech. V závěrečné části práce pak následuje popis neurodegenerativních onemocnění, která souvisí s autofagií.

Abstract:

This bachelor thesis is describing the process of autophagy including mechanisms of its induction and detection. The first part of the thesis is dedicated to the proteins and protein complexes that are essential for this process. In the main part the possibilities of the autophagy focusing on \kur{in vitro} systems are introduced. Biological and chemical ways of induction including lithium, trehalose and metformin are summarized. In the next part of the thesis the methods of autophagy detection such as transmission electron microscopy, selected types of immunoanalysis and image-based cytometry are characterized in \kur{in vitro} systems. In the final part of the thesis the neurodegenerative diseases connected with autophagy are described.
