The importance of leptin in haematology and oncology

Abstract:

Leptin je malý 16kD protein s regulačními funkcemi hormonu a cytokinu. Tyto funkce jsou velmi různorodé. Hlavní funkcí je restrikce příjmu potravy prostřednictvím hypothalamických mediátorů. Leptin se však podílí na mnoha dalších funkcích včetně kontroly pohlavního vývoje a rovněž regulace krvetvorby a něktarých imunologických parametrů. Cílem práce bylo: 1) zjistit, zda existuje vztah mezi koncentracemi leptinu v séru a tukem v kostní dřeni, stejně jako je tomu u periferní tukové tkáně a viscerálního tuku,2)zjistit, zda existuje vztah mezi leptinem a parametry hematopoézy in vivo 3)zjistit nové poznatky o metabolizmu leptinu u pacientů s nádorovým onemocněním 4) zjistit případné odlišnosti v metabolizmu leptinu u nádorových onemocnění ve srovnání s nenádorovými onemocněními. Studie prokázaly několik dříve nepublikovaných faktů, zejména: 1)nižší koncentrace leptinu v kostní dřeni v porovnání s periferní krvi, 2) vzájemnou závislost těchto koncentrací, 3) závislost koncentrací leptinu v periferní krvi i v kostní dřeni s množstvím tukové tkáně v kostní dřeni 4) korelaci koncentrací leptinu v periferní krvi i kostní dřeni se stupněm infiltrace kostní dřeně nádorem u hematologických malignit, 5)zachovaný diurnální rytmus leptinu u pacientů se solidními nádory. 6) snížení sekrece leptinu podkožní tukovou tkání u některých solidních nádorů 7)absenci vztahu koncentrací leptinu v periferní krvi i kostní dřeni s množstvím erytroidních či myeloidních buněk ve dřeni Dále bylo zjištěno v souladu s některými literárními daty: 1)snížení koncentrací sérového leptinu u některých solidních nádorů, 2)ztrátu závislosti koncentrací leptinu a BMI a ztrátu diference v koncentracích leptinu mezi pohlavími u těchto pacientů.

Abstract:

Leptin is a small 16kD protein produced by adipose tissue. It has a regulatory function of hormone and cytokine. These functions are very polymorphic. The main function is the restriction of feeding through some mediators from hypothalamus. However leptin is concerned of many other functions including control of sex development, and also regulation of hematopoesis and some immunological functions. Aims: 1) Identify if relation between concentrations of leptin in serum and fat in bone marrow exists as well as in case of subcutaneous and visceral fat tissue, 2) ascertain if approach among leptin and parameters of hematopoesis in vivo exist, 3) uncover a new peaces of knowledge of leptin metabolism in cancer patients, 4) find appropriate differences in metabolism of leptin in tumour diseases compared with non-tumour disorders. Conclusions: The studies demonstrate a few previously unpublished facts especially: 1) lower concentrations of leptin in bone marrow compared to peripheral blood, 2) the dependence between these concentrations, 3) the dependence leptin levels in bone marrow and peripheral blood amount of the fat in bone marrow, 4) correlation of leptin concentrations in peripheral blood and bone marrow to the grade of infiltration of bone marrow by tumour in haematological malignancies, 5) preserved diurnal rhythm of leptin of patients with solid tumours, 6) reduction of secretion of leptin by subcutaneous fat tissue in some solid tumours,7)no relation of concentrations of leptin in peripheral blood and bone marrow to amount of erythroid and myeloid cells in bone marrow. So far in agreement with some data in literature was investigated: 1) the reduction of concentrations of leptin in serum of some patients with solid tumours, 2) lack of dependence of leptin concentrations to BMI and lack of difference of leptin concentrations between sexes of these patients.
