Comet assay: new trends and application in genotoxicity testing

Abstract:

Denně lidé přicházejí do styku s mnoha chemickými látkami a mnohdy si ani neuvědomují, jaký vliv mohou mít tyto látky na jejich zdraví. Proto je nutné zabývat se testováním jejich potenciálních toxických efektů. V této bakalářské práci je uveden a diskutován souhrn nejnovějších trendů v používání metody testovaní genotoxicity chemických látek známé jako Comet assay, srovnání softwaru používaného k jejímu vyhodnocení a optimalizace standardního operačního postupu. Výstupem je taktéž zavedení této optimalizované metody do laboratoří RECETOX. Cílem praktické části je pak stanovení genotoxicity ethylhexylpara-methoxycinamátu (EHMC), který je používán jako UV filtr v mnohých opalovacích krémech. Používání těchto opalovacích krémů vede k uvolňování EHMC do vod. Proto je v práci zahrnut i seznam možných reakcí a tedy i produktů vznikajících z EHMC. U těchto produktů je pak diskutována i jejich potenciální toxicita.

Abstract:

People are daily in contact with many chemicals and they often do not realize how these chemicals can affect their health. Therefore it is necessary to consider the testing of their potential effects. In this thesis is presented and discussed summary of latest trends in using the genotoxicity testing method known as Comet assay, also comparison of the software used to its evaluation and optimization of the standard operation procedure. Output is also introduction of this optimized method to RECETOX laboratories. Aim of the practical part is to test genotoxicity of ethylhexylparamethoxycinnamate (EHMC), which is used as UV filter in many sunscreens. Using these sunscreens leads to release the EHMC to waters. Therefore, this work also includes the list of possible reactions of EHMC and their products. Potential toxicity of these products is discussed.
