Structure investigation of hydrogels using a cryo-SEM

Abstract:

Gely jsou materiály s unikátními vlastnostmi, které v sobě snoubí prvky jak pevných látek, tak i látek kapalných. Jsou tvořeny pevnou sítí a v ní zachycenou kapalinou. Gely, které obsahují vodu, se nazývají hydrogely a mohou najít uplatnění v mnoha oborech, z nichž nejvýznamnějšími jsou lékařství, kosmetika a farmacie. Tato práce se zabývá zkoumáním struktury hydrogelu kyseliny hyaluronové a agarózového hydrogelu a její závislosti na koncentraci, a to pomocí metod mrazové fixace a zobrazování v cryo SEMu. Zároveň jsou nalezeny a popsány nejvhodnější techniky přípravy a mražení.

Abstract:

Gels are materials with unique properties combining features of both solids and liquids. They form a solid network with a liquid trapped inside. Gels incorporating water are called hydrogels and they can be applied in many fields, most importantly medicine, cosmetics and pharmacy. Within this thesis, the hyaluronic acid hydrogel and agarose hydrogel structures and its dependence on concentration is examined using cryo fixation and cryo SEM imaging, and the most suitable preparation and freezing methods are found and described.
