Molekulární fylogeneze a evoluce neotenie v nadčeledi Elateroidea (Coleoptera)

Molecular phylogeny and evolution of neoteny in the superfamily Elateroidea (Coleoptera)

Abstract:

Elateroidea sensu lato je druhově nejpočetnější skupinou série Elateriformia a šestou největší nadčeledí Polyphaga. Obsahuje na jedné straně linie se silně sklerotizovaným tělem a využívající klikací mechanismus jako únikovou reakci před predátory (např. Elateridae), na druhé straně také linie měkkotělé, s častou přítomností neotenických forem, chemické obrany, aposematického zbarvení a bioluminiscence (Lampyridae, Cantharidae). Tyto na první pohled morfologicky rozdílné skupiny byly v minulosti řazeny do oddělených nadčeledí Elateroidea sensu stricto a Cantharoidea a teprve nedávno byly spojeny do společné nadčeledi. Fylogenetické vztahy mezi jednotlivými liniemi uvnitř Elateroidea jakožto i otázka jejich pozice v rámci série Elateriformia však dosud nebyly uspokojivě vysvětleny. Abych prozkoumal pozici Elateroidea v systému Coleoptera a poskytl hypotézu o příbuzenských vztazích mezi elateroidními liniemi, sekvenoval jsem dva ribozomální (18S, 28S) a dva mitochondriální (16S, COI) molekulární markery téměř 300 elateriformních zástupců. Nové sekvence jsem spojil s dosud nepublikovanými daty vytvořenými v Laboratoři molekulární fylogenetiky PřF UP a daty volně dostupnými z databáze GenBank. Počet terminálních linií Elateroidea v datových souborech byl několikanásobně vyšší než v předchozích studiích a tato práce se tak stala nejobsáhlejší analýzou fylogenetických vztahů v rámci zkoumané nadčeledi. Finální datové soubory byly alignovány několika programy a analyzovány metodami maximální parsimonie, maximální věrohodnosti a bayesiánské inference. Výsledky mých analýz dokazují vhodnost zvolených markerů pro fylogenetické studie na úrovni vyšších taxonů a v mnoha ohledech se shodují s nejnovějšími molekulárně i morfologicky založenými studiemi o fylogenezi Coleoptera. Potvrdil jsem monofylii Elateriformia, široce definovaných Elateroidea v sesterské pozici vůči Byrrhoidea i většiny analyzovaných čeledí vyjma Elateridae a Omethidae. Uvnitř Elateridae se ve všech analýzách jako terminální linie Agrypninae objevili měkkotělí Drilidae a jejich status byl snížen z čeledi na tribus. V čeledi Omethidae byli ve většině analýz přítomni Telegeusidae a jejich status byl snížen na úroveň podčeledi. Výsledky mé práce nepotvrdily blízkou příbuznost silně sklerotizovaných čeledí Elateridae, Eucnemidae, Throscidae a Cerophytidae ani monofylii bývalých cantharoidních linií a potvrdily hypotézu o mnohonásobném vzniku měkkého těla, neotenie a bioluminiscence v rámci nadčeledi. Vysoce podpořen byl blízký vztah Lampyridae a Cantharidae a konzistentně napříč analýzami byl přítomen klád Phengodidae + Rhagophthalmidae + Omalisidae + Elateridae. Dřívější hypotéza o příbuznosti Rhagophthalmidae a Lampyridae byla tedy odmítnuta. V alfa-taxonomické části mé práce jsem se zaměřil na studium neotenické čeledi Drilidae (nyní tribus Drilini), jejíž klasifikace je založena na historických pracích a je dodnes v chaotickém stavu, což znemožňuje využití této skupiny v evolučně orientovaných studiích. Zaměřil jsem se na pozici rodu Pseudothilmanus, který byl v literatuře zmíněn naposled roku 1944 v katalogu čeledi Drilidae. Od té doby tento rod nebyl studován a zůstal v nejistém postavení až než byl v rámci této práce na základě studia externích morfologických znaků převeden do Rhagophthalmidae. Review himalájských druhů rodu Selasia přineslo dva nové druhy a jiný druh byl naopak přesunut do Lampyridae. Při studiu fauny Drilini ze Sokotry jsem popsal další druh nový pro vědu a poukázal na fakt, že dřívě popsaný druh by měl být na základě odlišné vnější morfologie i samčích pohlavních orgánů vyřazen z tribu Drilini. Abstract:

Elateroidea sensu lato is the most speciose group of series Elateriformia and sixth largest superfamily of Polyphaga. There are two morphologically different groups of beetles in Elateroidea - the first assemblage with fully sclerotized body and clicking mechanism as an anti-predatory strategy (e.g. Elateridae) and second one with soft body and often containing neotenic incompletely metamorphosed forms, using chemical defense, aposematic coloration or bioluminescence as antipredator adaptations (Lampyridae, Cantharidae). These groups were treated as independent superfamilies Elateroidea sensu stricto and Cantharoidea and have been merged together only recently. Phylogenetic relationships among elateroid lineages as well as the position of Elateroidea within Elateriformia remain basically unresolved and are still under debate. To investigate the phylogeny of Elateroidea and the position of elateroid lineages within Coleoptera, I sequenced two ribosomal (18S, 28S) and two mitochondrial (16S, COI) molecular markers of almost 300 elateriform individuals. The newly sequenced data were merged with unpublished sequences produced in the Laboratory of Molecular Phylogenetics (UPOL) and sequences downloaded from GenBank. The number of elateroid terminals in newly produced datasets is severalfold higher than in previous analyses and, therefore, this study is the most comprehensive hypothesis of elateroid phylogeny ever. Final matrices were aligned using several algorithms and analyzed by maximum parsimony, maximum likelihood and Bayesian approaches. The results of the phylogenetic analyses confirmed the used genes as appropriate for study of family level relationships, and are in agreement with the recent molecular and morphology-based phylogenies of Coleoptera. My results confirmed the monophyly of Elateriformia, wide definition of Elateroidea, sister relationships of Elateroidea and Byrrhoidea, and monophyly of the most analyzed families except Elateridae a Omethidae. Softbodied family Drilidae was consistently found as an internal lineage within Elateridae: Agrypninae and, therefore, its status was lowered to tribe. In most of the analyses, Telegeusidae were found within Omethidae and were lowered to subfamily rank. The close relationships of fully sclerotized families Elateridae, Eucnemidae, Throscidae and Cerophytidae as well as monophyly of former Cantharoidea were rejected. The results of the phylogenetic analyses confirmed the hypothesis of multiple origin of softbodiedness, neoteny and bioluminescence within Elateroidea. The Lampyridae and Cantharidae were well-supported as sister groups. The results show the consistent support of Phengodidae + Rhagophthalmidae + Omalisidae + Elateridae clade so the previously published hypothesis on the close relationships of Rhagophthalmidae and Lampyridae was rejected. In alpha-taxonomical part of my thesis I focused on the study of neotenic family Drilidae (recently tribe Drilini). The classification of the group is based only on historical studies and is absolutely chaotic to date which preclude the usage of this lineage in evolutionary oriented research. Genus Pseudothilmanus, which was catalogued in Drilidae in 1944 but has never been studied again was transferred to Rhagophthalmidae on the basis of similar external morphology. In review of the Himalayan species of Selasia I described two species new for science and transferred one to Lampyridae. During my study of the Drilini fauna of Socotra Island I described one new species and discussed the possibility that previously described Socotran species should be excluded from Drilini after the detailed investigation of external morphology and male genitalia. Therefore, this thesis does not only give us the robust hypothesis of Elateroidea phylogeny and confirms the multiple origin of neoteny within elateroid lineages but also helps to improve the classification of Drilini - the group interesting from an evolutionary point of view.
