Biofilm Formation on Polymer Surfaces

Abstract:

V bakalářské práci je popsán vztah mezi polymerními povrchy a tvorbou biofilmu plísní. V teoretické části jsou shrnuty informace o biomateriálech, biofilmu a povrchové energii, která byla měřena u všech vzorků. V experimentální části je zkoumána tvorba biofilmu plísní na různých polymerních nátěrech. Zkoumané vzorky polymerních nátěrů byly na bázi epoxyesterové a methylsilikonové pryskyřice. V experimentu byly využity plísně Aspergilus niger, Trichoderma viride a Paecilomyces variotii. Z těchto plísní byla vytvo-řena směsná kultura, která byla naočkována na vzorky. Bylo sledováno, jak je biofilm schopen přerůstat vzorek anebo tvořit biofilm přímo na něm. Kultivace probíhala na kompletním a nekompletním agaru. Pro stanovení povrchové energie byla použita metoda měření kontaktního úhlu sedící kapkou. Dále bylo testováno, zda je biofilm schopen čerpat živiny pouze z živné půdy nebo i ze samotného vzorku. Hodnoty růstu a přerůstání byly dány do vztahu ke složení vzorků a k naměřeným hodnotám povrchové energie.

Abstract:

The bachelor thesis describes the relationship between polymer surfaces and fungal biofilm. The theoretical part summarizes the information about biomaterials, biofilm and surface energy, which were measured for all samples. In the experimental part the forma-tion of fungal biofilm on different polymer coats is investigated. The investigated samples of polymer coatings were based on epoxy and methylsilicone resins. Aspergillus niger, Trichoderma viride and Paecilomyces variotii were used in the experiment. A mixed culture was formed from these fungi. The mixed culture was seeded on samples. The biofilm was able to overcome the sample or create a biofilm directly on it. Cultivation takes place on complete and incomplete agar. For the determination of surface energy, a method of measuring a contact angle was used. It has also been tested whether the fungi was able to get some nutrients from the sample itself. Growth and overgrowth values were determined and put into the relation with composition of the samples and the surface energy.
