Methods for repellency testing in ticks

Abstract:

Tato práce se zabývala testováním repelentů proti klíšťatům Ixodes ricinus za pomoci vybraných metod. Testovány byly běžně dostupné přípravky: Repelent sprej s účinnou látkou IR3535, ANTImoskyto s účinnou látkou DEET a Zeckito s účinnou látkou icaridin. Jejich repelentní účinnost byla ověřována třemi metodami: vertikálními testy, testy na Petriho miskách a pomocí Moving-object bioassay. Ve vertikálním testu a při využití metody Moving-object bioassay byly účinnější Repelent sprej a ANTImoskyto, oba odpudily 100 % klíšťat. V testu na Petriho miskách byl nejúčinnější ANTImoskyto repelent, který odpudil 96 % klíšťat. Přípravky Repelent sprej a ANTImoskyto byly statisticky významně účinnější než Zeckito, rozdíl mezi jejich repelentní účinností ovšem statisticky významný nebyl. Klíšťata pro testy byla sbírána v lokalitě Brno – Pisárky v průběhu roku 2011. Celkem bylo nasbíráno 762 klíšťat. Nejvíce bylo nymf (91 %), samic bylo 5 %, samců 3,7 % a larev 0,3 %. Současně byly zaznamenávány hodnoty teploty, vlhkosti a tlaku vzduchu, ale závislost aktivity klíšťat na těchto faktorech se nepodařilo prokázat. Po testech byla klíšťata vyšetřena mikroskopií v zástinu na přítomnost patogenních spirochét, které by mohly patřit mezi původce onemocnění lymeské boreliózy. Celková prevalence byla stanovena na 16,35 %, infekční byly pouze nymfy (18,18 %) a samice (11,11 %).

Abstract:

This work dealt with the testing of repellents against ticks Ixodus ricinus using selected methods. Products tested were all commercially available: Repelent sprej with active component IR3535, ANTImoskyto with active component DEET and Zeckito with active component icaridin. Their repellent efficacy was verified by three methods: vertical climbing assay, the Petri dish assay and using Moving-object bioassay. In the vertical climbing assay and Moving-object bioassay were more effective Repelent sprej and ANTImoskyto, both of them repelled 100 % of ticks. In the Petri dish assay was the most effective repellent ANTImoskyto which repelled 96 % of ticks. Products Repelent sprej nad ANTImoskyto were statistically significantly more effective than Zeckito but the difference between repellent efficacy of Repelent sprej and ANTImoskyto was not statistically significant. Ticks for these tests were collected in the locality Brno – Pisarky by flagging method during 2011. Totally 726 ticks were collected. Most of them were nymphs (91 %); females were 5 %, males 3.7 % and 0.3 % were larvae. At the same time were recorded the temperature, humidity and air pressure, however the tick activity dependence on these factors could not be demonstrated. Ticks were also examined by dark field microscopy for the presence of pathogenic spirochetes that could be among the causative agent of Lyme borreliosis. The overall prevalence was set at 16.35%, only nymphs (18.18 %) and females (11.11 %) were infectious.
