Genome-wide profile of copy number changes associated with progression in monoclonal gammopathies

Abstract:

In this thesis we study the progression of monoclonal gammopathy of undetermined significance and asymptomatic multiple myeloma to the stage of multiple myeloma. Multiple myeloma is a genetic disorder, where the most commonly observed abnormalities are copy number variations and loss of heterozygosity (especially uniparental disomy). However, a genetic abnormality, which would sufficiently predict the progression of the disease, has not yet been identified. Understanding the process of the disease progression could result in application of new treatments, which would slow down or preferably completely stop the process of progression. This bachelor thesis provides an overview of the genetic changes observed in the developing stages of monoclonal gammopathies. Other sections of this thesis are dedicated to the evaluation of microarray data (SNP array). The workflow of microarray experiment, data preparation in Genotyping console and statistical processing are described in detail. In the applied section of this thesis the association of copy number variations and the association of uniparental disomy are evaluated using publicly accessible datasets.

Abstract:

V tejto bakalárskej práci sa venujeme progresii monoklonálnej gamapatie nejasného významu a asymptomatického mnohopočetného myelómu do štádia mnohopočetného myelómu. Mnohopočetný myelóm je genetické ochorenie, a najčastejšie preukázané abnormality sú zmeny počtu kópií génov a strata heterozygozyty (najmä uniparentálna dizómia). Doposiaľ však nebola objavená genetická abnormalita, ktorá by s istotou predikovala progresiu ochorenia. Porozumenie procesu progresie ochorenia by v budúcnosti mohlo viesť k zavedeniu nových liečebných postupov, ktorými by bol proces progresie spomalený, ideálne celkom zastavený. Bakalárska práca poskytuje prehlaď genetických zmien pozorovaných vo vývojových štádiách monoklonálnych gamapatií. Zvyšná časť práce je venovaná vyhodnoteniu dát z mikročipových technológií (SNP čip). Podrobne je opísaný postup mikročipového experimentu a príprava dát na štatistické spracovanie v programe Genotyping console. Pomocou využitia verejne dostupných dát je v rámci praktickej časti práce vyhodnotená asociácia počtu kópií génov a asociácia uniparentálnej dizómie.
