Analytical procedure of aminophenol S-conjugates

Abstract:

Paracetamol je široce používané analgetikum a antipyretikum a v doporučených dávkách je považován za bezpečný lék. Eliminace terapeutických dávek paracetamolu probíhá v játrech třemi hlavními cestami. Přibližně 80 % paracetamolu je konjugováno s kyselinou glukuronovou a 10 % paracetamolu je konjugováno se sulfátem. Zbylých 10 % je oxidováno pomocí CYP450 za vzniku N-acetyl-p-benzochinoniminu, který je dále metabolizován. Vzniká tak postupně konjugát paracetamolu s glutathionem, cysteinem a N-acetyl-L-cysteinem. Paracetamol a jeho výše zmíněné metabolity lze stanovit s využitím velkého množství metod. Jednou z nejčastěji používaných metod je v tomto případě vysokoúčinná kapalinová chromatografie s UV detekcí. Hlavním cílem této práce bylo optimalizovat stanovení paracetamolu a jeho metabolitů pomocí vysokoúčinné kapalinové chromatografie s UV a MS detekcí. Námi získané meze detekce byly výrazně nižší s využitím MS. Dalším cílem této práce bylo pomocí preparativní kapalinové chromatografie purifikovat konjugát paracetamolu s glutathionem, cysteinem a N-acetyl-L-cysteinem.

Abstract:

Paracetamol is a widely used analgesic and antipyretic drug that is considered to be safe at therapeutic doses. Elimination of therapeutic doses of paracetamol occurs in the liver by three major pathways: approximately 80% of the dose is metabolized by conjugation with glucuronic acid and 10% by conjugation with sulfate. The remaining part of the dose, i.e. 10%, is oxidized by CYP450 to N-acetyl-p-benzoquinone imine which is further conjugated with glutathione to form glutathione and then cysteine and N-acetyl-L-cysteine conjugates. Paracetamol and its metabolites can be determined by a number of methods. High Performance Liquid Chromatography linked with UV detection is one of the most commonly used methods. The main aim of this thesis was to optimize the determination of paracetamol and its metabolites by HPLC linked with UV or MS detection. The obtained detection limits were significantly lower using MS detection. Another aim of this work was to purify the glutathione, cysteine and N-acetyl-cysteine conjugates of paracetamol by the preparative liquid chromatography.
