Structure-function relationship of telomeric proteins

Abstract:

Tato diplomová práce se věnuje studiu interakcí vybraných proteinů shelterinového komplexu pomocí NMR spektroskopie, fluorescenční anizotropie a gelové retardační analýzy. Telomery jsou nukleoproteinové struktury vyskytující se na koncích chromozomů eukaryotických buněk. Telomerová DNA je tvořena z mnoha repetic konzervované sekvence – u člověka 5‘ TTAGGG-3’. Délka repetic se při každém dělení zkracuje. Telomerová DNA je prodlužována telomerázou, která je přirozeně aktivní v kmenových, embryonálních a také nádorových buňkách. U somatických buněk je telomeráza neaktivní nebo umlčena. Telomery jsou chráněny shelterinovým komplexem sestávajícím ze šesti základních proteinů. Tento komplex plní několik funkcí. Shelterin rozpoznává, váže, formuje a chrání telomerovou DNA. Shelterin reguluje přístup a aktivitu telomerázy. Centrálním proteinem shelterinu, který váže dvouřetězcovou DNA je TRF2. TRF2 tvoří homodimery, které tvoří a stabilizují telomerovou smyčku. Dimer TPP1-POT1 se váže na jednořetězcový přesah DNA. Cílem práce je objasnění vlivu N-koncové domény proteinu TRF2 na vazbu s telomerovou DNA a studium interakcí proteinů POT1 a TPP1. Nejdříve byla studována vazba bazické domény TRF2 na telomerovou DNA pomocí NMR metod za účelem získání informací o struktuře domény a jejich změnách během vazby. Dále byly připraveny zkrácené varianty proteinů POT1 a TPP1 pro bližší pochopení mechanizmu vazby těchto proteinů shelterinu. Interakce purifikovaných proteinů byly sledovány za využití metod fluorescenční anizotropie a gelové retardační analýzy. Podrobné znalosti o těchto interakcích mohou být využity při cílenému návrhu inhibitorů, které by vedly k účinnější léčbě nádorových onemocnění.

Abstract:

This diploma thesis focuses on studying interactions of shelterin proteins using NMR spectroscopy, fluorescence anisotropy and gel retardation analysis. Telomeres are nucleoprotein structures located at the end of chromosomes of eukaryotic cells. Telomeric DNA consists of repetitive sequence – for example 5'-TTAGGG-3' in humans. The length of telomeres is shortened at each cell division. Telomeric DNA is extended by telomerase, which is natively active in stem and embryonic cells. Telomerase is additionally activated in tumor cells. Telomerase is inactivated or silenced in somatic cells. Telomeres are protected by shelterin complex that consists of six proteins. Shelterin fulfills several functions. Shelterin proteins recognize, bind, fold and protect telomeric DNA. Additionally, shelterin regulates access and activity of telomerase. TRF2 is a central shelterin protein that binds double-stranded DNA. TRF2 forms homodimers, which fold and stabilize telomeric loops. TPP1-POT1 dimer binds to single-stranded DNA overhang. The main objectives of this study were to (i) clarify the effect of the N-terminal domain of the protein TRF2 on its binding to telomeric DNA (ii) study interactions between proteins POT1 and TPP1. Firstly, the binding of the basic domain TRF2 to telomeric DNA was described using NMR methods to obtain information on the structure of domains and its changes during detention. Secondly, POT1 and TPP1 proteins were prepared to understand the mechanism of their binding. Interactions of purified proteins were studied using methods of fluorescence anisotropy and gel retardation analysis. Achieved finding about of interactions within shelterin can be used to design targeted inhibitors that might lead to more effective treatments of cancers.
