Spectrophotometric and calorimetric study of frozen aqueous solutions

Abstract:

Mrazení roztoků je v praxi běžně užíváno v potravinářském a farmaceutickém průmyslu, proto je důležité znát, a umět popsat změny chování roztoků při tomto procesu. Tato diplomová práce se zabývá spektrofotometrickým a kalorimetrickým studiem zmražených roztoků, chlazených při různých rychlostech, neboť tato problematika doposud není zevrubně popsána. Je známo, že nečistoty ve vodných roztocích se při mražení zkoncentrují do žil mezi krystaly hexagonálního ledu, proto byla tato studie zaměřena na samotné žíly. Proces chlazení a ohřevu vodných roztoků NaCl byl pozorován diferenční skenovací kalorimetrií, zatímco změny polarity a pH vodných roztoků NaCl byly zkoumány UV-Vis absorpční spektrometrií, s použitím barviv jako indikátorů vlastností pozorovaných roztoků. Podle výsledků kalorimetrické studie docházelo při rychlém chlazení k vitrifikaci koncentrovaného roztoku v žilách, přičemž vypočítané parametry polarity (E_T^N) těchto koncentrovaných roztoků tento fakt podporují. Kromě toho analýza absorpčních spekter acidobazického indikátoru potvrdila inkorporaci iontů do mřížky ledu během chlazení.

Abstract:

The purpose of the thesis was to explore the not yet fully understood behaviour of frozen solutions. It is important to know and describe this behaviour, since freezing of solutions is a common praxis in the food and pharmaceutical industry. This diploma thesis deals with the spectrophotometric and calorimetric study of solutions cooled down at various cooling rates. Due to the fact that the impurities in aqueous solutions concentrate into veins upon freezing, the focus of the study was on the veins themselves. The cooling and heating process of aqueous NaCl solutions was observed by differential scanning calorimetry, while the changes of polarity and pH of NaCl solutions were examined by UV-Vis absorption spectrometry using dyes as indicators of mentioned physical-chemical properties of observed solutions. The calorimetric study shows the vitrification of concentrated solution in veins. Also, the computed polarity parameters (E_T^N) of the concentrated solutions in the veins supported the fact of vitrification. Moreover, the ion incorporation during cooling was proved after analysis of the absorption spectra of the acid-base indicator.
