Abstract:

Cukerné alkoholy jsou přírodní látky sladké chuti, s nižší sladivostí vzhledem k sacharose. Jsou metabolizovány v tlustém střevě (erythritol v tenkém střevě), proto mají nízkou energetickou hodnotu. Nejsou úplně stravitelné, nestrávená část slouží jako vláknina. Nezpůsobují zubní kaz. Jsou vhodné pro diabetiky. Erythritol jako jediný nemá projímavé účinky. Xylitol chrání zuby před zubním kazem, neboť inhibuje růst bakterie Streptococcus mutans, zodpovědné za vznik zubního kazu. Cílem práce bylo pokusit se vyrobit xylitol ze slámy, stanovit co nejlepší podmínky a metody detekce. Dále pokusit se ověřit účinek xylitolu na S.mutans. Xylitol byl vyroben kyselou hydrolýzou pšeničné slámy a následnou redukcí hydrolyzátu kvasinkami Pichia quilliermondii (redukují xylosu na xylitol). Jako detekční metoda nejlépe posloužila vysoce účinná kapalinová chromatografie s hmotnostní detekcí. Bylo prokázáno, že xylitol ve vzorcích obsažen byl. Kvůli velkému množství sodíku, ale nebylo možné provést kvantifikaci. Také bylo potvrzeno, že xylitol značně omezuje růst bakterie Streptococcus mutans.

Abstract:

Sugar alcohols are natural substances taste sweet, with lower sweeter in reference to sucrose. They are metabolised in the colon (erythritol in the small intestine), therefore have a low energy value. They are not completely digested, undigested part serves as a fiber. Does not cause tooth decay. They are suitable for diabetics. Only erythritol is not a laxative effect. Xylitol protects teeth against dental caries, it inhibits the growth of the bacteria Streptococcus mutans, responsible for tooth decay. The target of this thesis was to try produce xylitol from straw, to determine the best conditions and methods of detection. Next try to verify the effect of xylitol on S.mutans. Xylitol was produced by acid hydrolysis of wheat straw and subsequent reduction this hydrolysate by Pichia quilliermondii (reduces xylose to xylitol). The best detection method was high-performance liquid chromatography with mass spectroscopy. It was shown that xylitol was included in the samples. It was impossible to quantify, due to the large amounts of sodium. It was also confirmed that xylitol inhibits the growth of bacteria S.mutans.
