Identification of proteins secreted by Salmonella

Abstract:

Bakterie rodu Salmonella jsou gramnegativní, fakultativně anaerobní tyčky, které se vyznačují ve většině případů pohyblivostí a schopností intracelulárního přežívání. Tento intracelulární patogen, který v závislosti na serovaru a hostitelském organismu, je schopen způsobit nemoci méně závažné jako gastroenteritidy, ale také závažné onemocnění břišní tyfus. Příznaky onemocnění jako horečka, průjem, zvracení a nevolnost jsou způsobeny jednak reakcemi imunitního systému hostitele, a jedna také efektorovými molekulami, které bakterie secernuje do hostitelských buněkTěmito molekulami jsou proteiny, které jsou kódovány na diskrétních ostrovech a ostrůvcích patogenity, podle nichž také dostaly jméno - ostrovy patogenity – SPI – Salmonella Pathogenicity Islands. Tyto proteiny jak přímo, tak nepřímo stimulují signální dráhy hostitele, což následně vede k různým odpovědím ze strany hostitelské buňky. Salmonella enterica využívá dva funkčně odlišné sekreční systémy III. typu, které jsou kódovány na ostrovech patogenity SPI-1 a SPI-2. Hlavní funkcí sekrečního systému SPI-1 TTSS-1 je umožnit bakteriální invazi do epiteliálních buněk. Druhý sekreční systém III. typu kódovaný SPI-2 TTSS-2 má za funkci usnadnění replikace intracelulárně umístěné bakterie uvnitř vakuoly SCV. V této práci jsem identifikovala proteiny, které do prostředí secernují salmonely. Secernované proteiny delečních mutantů S. Enteritidis a serovarů S. enterica z média byly sráženy pomocí kyseliny trichloroctové a kvantifikovány pomocí spektrofotometrie. Byly zavedeny techniky polyakrylamidové elektroforézy v přítomnosti dodecylsíranu sodného, které umožnily jejich vizualizaci. Vybrané separované proteiny byly podrobeny fragmetační analýze pomocí hmotnostní spektrometrie MALDI/TOF TOF MS a následně identifikovány. Většina proteinů patřila do dvou funkčních skupin – proteiny sekrečního systému III. typu a flagelárního aparátu.

Abstract:

Salmonella spp. are Gram-negative, rod shaped, mostly motile, facultative anaerobes, with ability to survive intracellulary. Based on serovar, these pathogenic bacteria can cause mild forms of infections - gastroenteritis, or can cause severe abdominal infections, like typhoid fever. Among symptoms belong fever, diarrhea, vomiting and nausea. The symptoms are caused by the immunoreactions of the host and by the interactions of the effecter molecules secreted by the bacteria. The genes located on discrete pathogenic islands and islets (SPI) code these effector molecules. The molecules stimulate signal pathways of the host. Either it can proceed directly or indirectly, which lead to different host immune reactions. Salmonella enterica uses two functional different secretory systems type III (TTSS), coded by SPI-1 and SPI-2. The main function of SPI-1 TTSS-1 is to enable the bacterium to invade the host epithelial cells. SPI-2 TTSS-2 has a function to facilitate the replication of the bacterium located in the SCV vacuole. Proteins secreted by Salmonella species to the environment were identified in this work. The proteins secreted by deletion mutants of S. Enteritidis and serovares S. enterica were precipitated by trichloracetic acid from a medium and subsequently were quantified by a spectrophotometry. SDS-PAGE technique was established with the aim to visualize these proteins. The selected proteins were analyzed by fragmentation on MALDI TOF/TOF mass spectrometry and subsequently were identified by comparison with databases. The most of the proteins belong to the two functional groups - proteins of the secretory system type III and the apparatus of flagella.
