Biogenesis and significance of sequence variants in microRNAs (isomiRs)

Abstract:

MikroRNA patří do skupiny krátkých nekódující RNA, které na post-transkripční úrovni regulují genovou expresi a významně se tak podílejí na regulaci klíčových buněčných procesů. Podstatná je také jejich role při vzniku a progresi mnohých onemocnění. V současné době je věnována pozornost diagnostickému potenciálu mikroRNA a v rámci biomarkerových studií jsou analyzovány jejich expresní profily. Implementace metody sekvenování nové generace pro účely expresního profilování vedla ke zjištění, že jsou jednotlivé mikroRNA kromě kanonických variant anotovaných v databázi miRBase zastoupeny dalšími izoformami. Tyto sekvenční varianty (také zvané izomiRs) lišící se v primární struktuře mohou vykazovat vyšší zastoupení než kanonické mikroRNA, mít odlišné biologické vlastnosti a zásadně ovlivňovat výsledky biomarkerových studií. Recentní práce také poukazují na jejich možný klinický význam.

Abstract:

MicroRNAs are a class of small non-coding RNAs which regulate gene expression at the post-transcriptional level and are significantly involved in the regulation of key cellular processes. They play an important role in the development of many diseases. Currently, the research of microRNAs is focused on diagnostic potential of these molecules and their expression profiles are analysed in many biomarker studies. Implementation of next-generation sequencing for the purpose of expression profiling have led to a finding that individual microRNAs are represented by other isoforms in addition to the canonical variants annotated in the miRBase database. These sequence variants (also called isomiRs), which differ from the canonical variants in the primary structure, can be more abundant, have different biological properties and influence the results of biomarker studies. Recent publications also suggest their potential clinical significance.
