Species delimitation, ecology and distribution of Fomes fomentarius and Fomes inzengae in the South Moravia region

Druhové rozlišení, ekologie a rozšíření Fomes fomentarius a F. inzengae v Jihomoravském kraji

Abstract:

Fomes fomentarius a Fomes inzengae jsou blízce příbuzné druhy hub bílé hniloby a jsou považovány za kryptické druhy. Morfologické znaky obou druhů jsou podobné a jejich rozlišení je obtížné. Tato práce si klade za cíl vymezení těchto druhů na základě mikroskopických a makroskopických morfologických znaků plodnic a multigenové fylogenetické analýzy sekvencí DNA jedinců rostoucích na různých dřevinných hostitelích v různých zeměpisných oblastech České republiky, především na území Jihomoravského kraje. Soubor dat obsahoval 48 položek rostoucích na 12 rodech hostitelských dřevin v České republice a na Slovensku. Analýza počtu rourek na jeden centimeter, doporučovaného jako rozlišovací znak obou druhů v literatuře, odhalila, že tyto hodnoty se u obou druhů překrývají, i když rozdíly vyšly jako statisticky signifikantní. Plodnice druhu F. fomentarius mají 10-28 pórů/cm a rozměry basidiospor 10,3‒22,9 × 4,8‒10,3 μm. F. inzengae má 12-31 pórů/cm; a rozměry basidiospor 10,8‒22,6 ×4‒9,8 μm. Fylogenetické studie analyzovaly soubor dat čtyř lokusů: Vnitřní transkribovanou oblast mezerníků (ITS) a velkou podjednotku (LSU) jaderné ribozomální DNA a dvou geny kódující proteiny: Translační elongační faktor 1-alfa (TEF1) a první podjednotku genu pro RNA polymerázu II (RPB1). Bylo získáno 47 sekvencí ITS, 10 sekvencí LSU, 9 sekvencí TEF1, 10 sekvencí RPB1. Pro fylogenetické analýzy byly použity metody maximální věrohodnosti (ML) a Bayesovské (BY) fylogenetické analýzy. Fylogenetické analýzy ukázaly, že ITS, LSU, TEF1 a RPB1 poskytují podobnou topologii stromů a konzistentní závěry potvrzující fylogenetické vymezení obou druhů. Geografické rozšíření a preference hostitele těchto dvou druhů se liší – F. fomentarius se vyskytuje hlavně ve vyšších nadmořských výškách, F. inzengae je hojnější v nížinách. Ačkoli morfologické znaky nemohou pomoci při identifikaci těchto dvou druhů, preference a geografické distribuce podporují oddělení ekologických u odlišných druhů.

Abstract:

Fomes fomentarius and Fomes inzengae are white rot fungi and have been considered as cryptic species. The morphological characteristic of both species may similar in some features that so challenging to distinguish. Thus, the aim of this work was delimitation of both species base on morphological characters of the basidiocarps and multi-gene phylogenetic analysis of DNA sequences of the specimens growing on different woody hosts of different geographic regions in the Czech Republic, mainly in the South Moravian region. The dataset included 48 basidiomes from 12 host tree genera within Czech Republic and Slovakia. The values of number of pores per centimeter, recommended as discriminative character of the two species in literature, overlapped, although resulted as significantly different in statistical test. The basidiomes of F. fomentarius have 10-28 pores/cm; the basidiospore dimensions were 10.3‒22.9 × 4.8‒10.3 μm. F. inzengae have 12-31 pores/cm; the basidiospore dimensions were 10.8‒22.6 ×4‒9.8 μm. Phylogenetic analyses analyzed the of four loci data-set: The internal transcribed spacer region (ITS) and the large subunit (LSU) of the nuclear ribosomal DNA (rDNA) regions and two protein-coding genes: Translation elongation factor 1-alpha (TEF1) and the 1 subunit of RNA polymerase II gene (RPB1). This study generated 47 ITS sequences, 10 LSU sequences, 9 TEF1 sequences, and 10 RPB1 sequences. Maximum likelihood (ML) and Bayesian (BY) phylogenetic analyses were used for phylogenetic analyses. All phylogenetic analyses showed that ITS, LSU, TEF1, and RPB1 provide similar tree topology and consistent conclusions representing the delimitation of the two species. The geographic distribution and host preference of the two species differ F. fomentarius growths mainly in higher altitudes, while F. inzengae is dominant in lowlands. Although the morphological characters cannot help in the identification of the two species, preferences and geographic distributions support the separation of the ecological in distinct species.
