Preparation of recombinant AFL1 protein in yeasts

Abstract:

Na našem pracovišti bylo již v minulosti provedeno několik experimentů s lektiny z plísně Aspergillus fumigatus. Samotný gen afl1 byl klonovacími procesy vnesen do plasmidu, který byl následně natransformován do buněk Escherichia coli. Cílem bakalářské práce bylo pokusit se připravit vhodný konstrukt obsahující gen afl1, tak aby bylo možno produkovat rekombinantní protein AFL1 v eukaryotickém systému kvasinky Pichia pastoris. Byla ověřena přítomnost cílového genu ve vstupním materiálu a následně byl tento gen namnožen pomocí polymerasové řetězové reakce pomocí k tomu určených primerů. Reakce proběhla opakovaně úspěšně. Takto získaná DNA byla podrobena štěpení pomocí restrikčních endonukleas, stejně jako cílový plasmid pPICZaA. První pokusy o následnou ligaci vedoucí k přípravě konstruktu byly neuspokojující, a proto byly dále optimalizovány. Byla optimalizována čistota vstupní DNA, složení ligační směsi a proces transformace. Z hlediska čistoty vstupní DNA bylo využito přečištění DNA z roztoku po polymerasové řetězové reakci a rovněž po následném štěpení restrikčními endonukleasami. Cílový plasmid byl po štěpení rovněž přečišťován, tentokrát pomocí elektroforezy izolací z agarosového gelu. Ligace byla upravována využitím různých molárních poměrů a různých množství DNA. Vzhledem k nepříznivým výsledkům byla vyzkoušena i metoda In-Fusion, pro kterou byly speciálně navrženy primery a následně optimalizována PCR. Výsledky této metody však nebyly uspokojivé. Ve snaze o zvýšení transformační účinnosti byla namísto dosud prováděné metody teplotního šoku, která se prokázala jako neúspěšná, využita i metoda elektroporace. Zatím nebyly bohužel první pokusy úspěšné.

Abstract:

Lectins from Aspergillus fumigatus have been studied at our institut. The gene afl1 was cloned into a plasmid, which was subsequently introduced into Escherichia coli cells. The aim of this thesis was to prepare a suitable construct containing gene afl1, in order to produce the recombinant protein AFL1 in eukaryotic system of Pichia pastoris. The presence of the target gene in the input material was verified and then the gene was amplified using polymerase chain reaction and primers designed for this purpose. The reaction was repeatedly successfull. In this manner acquired DNA was subjected to cleavage by restriction endonucleases, as well as the target plasmid pPICZαA. The first experiments of consequential ligation leading to preparation of the construct were unsatisfactory, and therefore were further optimized. The purity of input DNA, composition of the ligation mixture and transformation process were modified. Concerning the purity of the the input DNA, DNA was purified from the solution after polymerase chain reaction and also after the subsequent cleavage by restriction endonucleases. The target plasmid was also purified after cleavage and isolated from agarose gel after electrophoresis. Ligation was modified by using of different molar ratios and different amounts of DNA. Due to negative results we tested also the In-Fusion method, for which we specially designed primers and optimized PCR consecutively. However the results of this method were not satisfactory. In an effort to increase the transformation efficiency, the electroporation method was used. Unfortunately, the first trials have not been successful, so far.
