Characterization of red clover candidate genes for biological nitrogen fixation and study of their polymorphisms

Abstract:

Symbiotická fixace dusíku je mutualistický vztah mezi rostlinou a bakterií. V tomto symbiotickém vztahu rostlina poskytuje bakterii zdroj uhlíku a energie a bakterie dodává rostlině dusík zabudovaný do organických sloučenin, které je rostlina schopná ve svém metabolismu využít. Nejznámější a nejprobádanější je symbiotický vztah mezi rostlinami čeledi Fabaceae a hlízkovými bakteriemi. Cílem této diplomové práce je identifikovat a charakterizovat klíčové geny jetele lučního (Trifolium pratense L.) podílející se na symbiotické fixaci vzdušného dusíku metodami bioinformatické a molekulárně biologické analýzy. Za pomoci celogenomových sekvencí a sekvencí získaných obohaceným sekvenováním budou hledány funkční rozdíly a polymorfismy v těchto kandidátních genech. V experimentální analýze budou využity kontrastní fenotypy jetele lučního pro symbiotickou fixaci dusíku pro provedení expresních analýz. Na základě exprese kandidátních genů bude ověřováno, zda jejich exprese koreluje s fenotypovými projevy, projevujícími se množstvím fixovaného dusíku.

Abstract:

Symbiotic nitrogen fixation is a mutualistic relationship between plant and bacteria. In this symbiotic relationship plant provides source of carbon and energy to bacteria and bacteria supplies plant with nitrogen converted into organic compounds which can be utilized by plant. The best known and best researched symbiotic relationship occurs between plants of the family Fabaceae and rhizobacteria. The aim of this diploma thesis is to identify and characterize red clover (Trifolium pratense L.) key genes participating in symbiotic nitrogen using methods of bioinformatic and molecular biology analyses. Functional differences and polymorphisms in these candidate genes will by sought in whole genome sequences and sequences acquired by target-enrichment sequencing. In experimental part of this thesis contrast phenotypes in symbiotic nitrogen fixation of red clover will be used to perform expressional analyses. Based on the expression of candidate genes, it will be verified whether their expression correlates with phenotypic manifestation of amount of fixed nitrogen.
