Determination of acetylcholinesterase activity in the earthworm Eisenia foetida after exposure to rock waste

Abstract:

Diplomová práce je zaměřená na problematiku dopadu těžby nerostných surovin na krajinu a stále více se rozvíjejícím biotestům. Úvodní kapitola se zabývá problematikou těžby nerostných surovin. Jsou zde uvedeny příklady výrazně pozměněných míst krajiny v celosvětovém měřítku. Dále je pozornost věnována místům v České republice, která byla výrazně narušena těžbou nerostných surovin. V jednotlivých podkapitolách jsou popsány způsoby těžby některých typů nerostných surovin v naší krajině a jejich vliv na okolní prostředí. Jsou zde uvedeny také vhodné postupy rekultivačních prací s ohledem na zachování nebo obnovu krajiny. Rešeršní část se ve své druhé polovině věnuje stále více se rozvíjejícím ekotoxikologickým testům. A hodnocení vlivu toxických nebo potencionálně nebezpečných látek pro živé organismy prostřednictvím specifických biomarkerům, které se jeví jako vhodný nástroj pro hodnocení toxicity. Hlavním cílem této práce bylo stanovit aktivitu enzymu acetylcholinesterézy po expozici horninovým odpadem. Jednalo se o lokalitu z odvalu dolu Jeremenko, ve které jakékoli dosavadní pokusy o zalesnění zcela selhaly. Aktivita enzymu AChE byla hodnocena také u jedinců po expozici s testovanou látkou Nurelle D. Tato látka patří do skupiny insekticidů, které výrazně ovlivňují aktivitu tohoto enzymu. Byly provedeny čtrnáctidenní testy akutní toxicity na modelovém organismu Eisenia foetida.

Abstract:

The thesis is focuses on the issue of the impact of mineral exploitation on the landscape and are increasingly developing biotests. The first chapter deals with the extraction of mineral resources. There are examples dramatically modified the landscape of places worldwide. Further attention is paid places in the Czech Republic, which was significantly disrupted mining of mineral resources. In the subsections are described methods for the extraction of certain types of mineral resources in our country and their impact on the environment.. The section also includes appropriate procedures the reclamation works for the conservation and restoration of the landscape. The review part in its second half focuses ever more developing ecotoxicological tests. And assessing the impact of toxic or potentially hazardous substances to living organisms using specific biomarkers that appear to be a useful tool for evaluation of toxicity. The main aim of this study was to determine the activity of the enzyme acetylcholinesterase after exposure to rock waste. It was the site of the mine dump Yeremenko in which any previous attempts at reforestation of the site completely failed. AChE enzyme activity was also assessed in individuals after exposure to the test substance Nurell D. This substance belongs to the group of insecticides, which significantly affect the activity of this enzyme. Were carried out fortnightly acute toxicity tests on model organisms Eisenia foetida.
