Vztah polymorfismu MHC genů a diverzity společenstev mnohobuněčných parazitů v populacích druhu Squalius cephalus (L.) z vybraných evropských povodí

The link between MHC polymorphism and diversity of metazoan parasite communities in populations of Squalius cephalus (L.) from selected European drainages

Abstract:

Studium hostitelsko-parazitických vztahů je úzce spjato s pochopením interakcí mezi hostiteli a parazity během relativně dlouhého evolučního a relativně krátkého ekologického času. Hlavními cíli předložené dizertační práce bylo (1) testování biogeografických hypotéz druhové diverzity mnohobuněčných parazitů, (2) studium fylogeografie hostitelského druhu (studium interakcí hostitel-parazit v prostorovém kontextu), (3) analýza struktury, diverzity a evolučních mechanizmů funkčních MHC IIB genů a (4) analýza vztahu mezi MHC IIB geny a parazitizmem (studium interakcí hostitel-parazit na molekulární úrovni). Modelovým hostitelem všech studií byla sladkovodní kaprovitá ryba jelec tloušť, Squalius cephalus (Linnaeus, 1758), charakterizovaná širokým areálem rozšíření, komplexní evoluční historií a bohatou parazitofaunou. Bylo analyzováno 15 populací jelce tlouště z téměř celého areálu jeho rozšíření v Evropě. Populace v okrajových oblastech distribuce jelce tlouště vykazovaly nižší hodnoty diverzity mnohobuněčných parazitů a rovněž nižší hodnoty genetické variability (mikrosatelity, cytochrom b a MHC geny). Analýza biogeografických gradientů diverzity prokázala vztah mezi zeměpisnou šířkou a diverzitou parazitů (hypotéza latitudinálního gradientu biodiverzity), pokles abundance parazitů směrem od příznivého centra k okrajovým oblastem výskytu parazitů (hypotéza modelu „příznivého centra“) a vliv fylogenetické a geografické vzdálenosti mezi populacemi na strukturu společenstev parazitů (hypotéza poklesu podobnosti se vzdáleností). Fylogeografické analýzy jelce tlouště v Evropě použitím jaderných i mitochodriálních markerů potvrdily platnost hypotéz předpokládajících existenci několika refugií během glaciálních období a rozdílné cesty kolonizace u sladkovodních evropských ryb. Analýza funkčních MHC IIB genů prokázala vysokou míru jejich polymorfizmu. Byla potvrzena přítomnost dvou MHC IIB lokusů (DAB1 a DAB3) a navíc možná duplikace DAB3 genu. Byl prokázán vliv pozitivní selekce a částečný podíl neutrální selekce na evoluci MHC genů, přítomnost rekombinace nebyla potvrzena. Fylogenetická analýza DAB genů potvrdila hypotézu mezidruhového polymorfizmu, na druhou stranu nebyl zaznamenán vztah mezi biogeografií a fylogenezí DAB alel. Vztah mezi MHC geny a parazitizmem byl potvrzen v souladu s hypotézou „výhoda heterozygota“ a hypotézou „na frekvenci závislá selekce“. Nicméně tento vztah byl výsledkem vlivu populace, který měl signifikantní podíl jak na variabilitu MHC tak i na diverzitu parazitů.

Abstract:

The studies of host parasite relationships are closely linked with the understanding how hosts and their parasites interact over relatively long evolutionary and relatively short ecological time. The main objectives of the thesis were: (1) testing biogeographical hypotheses of species diversity, (2) reconstruction of phylogeography of host species (biogeography of host-parasite interactions), (3) analysis of structure, diversity and evolutionary patterns of expressed MHC class IIB genes and (4) analysis of the link between MHC class IIB genes and parasitism (molecular host-parasite interactions). The chub (Squalius cephalus), a common European cyprinid species, was used as a model organism. A total of 15 populations of chub covering a wide range of its distribution in Europe were analyzed. The populations in marginal zones of chub distribution were characterized by low values of parasite diversity as well as by low values of genetic variability (microsatellites, cytochrome b and MHC genes). Analyses of biogeographical gradients of diversity revealed the correlation of parasite diversity with latitude (confirming the latitudinal gradient hypothesis), decrease of parasite abundance from locality with the most favourable conditions to the marginal zones of parasite distribution ('favourable centre' hypothesis) and a role of phylogenetic and geographic distance between host populations in the structure of parasite communities (distance decay in similarity). The phylogeographical analyses of chub populations support the hypotheses of the multiple-refuges origin and different routes of postglacial colonization of European freshwater fishes. The analyses of expressed MHC IIB genes revealed a high degree of MHC polymorphism in chub. The presence of two MHC class IIB loci (DAB1-like and DAB3-like) and possible duplication of DAB3 gene were confirmed. Testing the evolutionary mechanisms potentially driving the MHC diversity indicated that positive selection and partially also neutral selection can explain the MHC IIB evolution in chub, while recombination does not play an important role in MHC diversity. The phylogenetic analyses of DAB alleles did not indicate any biogeographical pattern of the genetic structure of MHC IIB, which is in line with balancing selection and trans-species polymorphism. A link between MHC genes and parasitism was confirmed in line with hypotheses of parasite-mediated balancing selection (heterozygote advantage and negative frequency-dependent selection). However, this relationship was confounded by population effect.
