Diversity of Resistant Bacteria and Study of Bacterial Resistome in Wastewater Treatment Plants and Distribution in the Environment

Abstract:

Wastewater treatment plants (WWTPs) are considered a potential reservoir of antibiotic-resistant bacteria (ARB). Horizontal transfer of the genetic information between different bacterial species, including the spread of antibiotic resistance genes (ARGs), can occur in the wastewater. The technology of treatment plants is not yet sufficient to achieve the complete elimination of antibiotic pollutants, and therefore there may be an increase in resistant strains in the wastewater. Local studies mapping antibiotic resistance make a significant contribution to gaining the necessary knowledge, which can be the starting point for incentives to reduce and stop its spread. The thesis detected the presence of ARGs in WWTPs during various phases of treatment processes and subsequently in the water recipient Žermanice dam in the Moravian-Silesian region of the Czech Republic. The effect of sub-inhibitory antibiotic concentrations on the bacterial biofilm formation was evaluated and resistant and multidrug-resistant bacterial strains were identified using molecular genetic methods. The results showed that ARGs were located in all studied tanks of WWTPs and also in the water recipient to which the water from the WWTP was drained. An interesting finding was that the sedimentation phase of the WWTP treatment was an ideal environment for the occurrence of ampicillin-resistant bacteria and for the accumulation of ARGs, especially the blaTEM and tetW genes. High-risk blaNDM-1 and vanA genes have also been detected in the wastewater. Although these genes belong to rare genes, they are classified as resistant genes to antibiotics of last resort. This is the first study confirming the presence of the blaNDM-1 gene in the Czech wastewater. Important findings also emerged from monitoring bacterial diversity in the wastewater. A shift was found between the occurrence of wild type bacteria and resistant bacteria in individual tanks of WWTPs. One of the most dominant taxa of ampicillin-resistant bacteria was the class Gammaproteobacteria, which may play an important role in the accumulation of resistance to beta-lactam antibiotics in the wastewater. Resistance isolates were identified, and resistance genes were confirmed. Co-resistance genes for beta-lactams, carbapenems and tetracyclines have been identified in multidrug-resistant strains. Aeromonas sp. dominated among the multidrug-resistant strains, which poses a certain danger due to the spread of resistance among different species of bacteria in the environment. The opportunistic pathogen Pseudomonas aeruginosa was also among the identified multidrug-resistant strains. Further research was focused on the effect of sub-inhibitory concentrations of the antibiotic tetracycline on the biofilm formation by resistant isolates from the wastewater. Tetracycline concentrations from 0.031 to 0.125 ?g/mL were found to have a stimulating effect not only on the bacterial growth but also on the bacterial biofilm formation. Resistant isolates forming a biofilm are a risk factor for the formation and spread of ARGs in the wastewater and further into the environment.

Čistírny odpadních vod (ČOV) jsou považovány za potenciální rezervoár antibioticky rezistentních bakterií (ARB). V odpadních vodách může docházet k horizontálnímu přenosu genetické informace mezi různými bakteriálními druhy včetně šíření antibioticky rezistentních genů (ARGs). Technologie čistíren není zatím dostačující pro dosažení úplné eliminace antibiotických polutantů, a proto v odpadních vodách může docházet k nárůstu rezistentních kmenů. Lokální studie mapující antibiotickou rezistenci významně přispívají k získání potřebných poznatků, které mohou být východiskem podnětů vedoucích k omezení a zastavení jejího šíření. V práci byla detekována přítomnost ARGs v odpadních vodách ČOV během různých fází čistících procesů a následně ve vodním recipientu Žermanická přehrada v Moravskoslezském kraji České republiky. Byl zhodnocen vliv sub-inhibičních koncentrací antibiotik na tvorbu bakteriálního biofilmu a identifikovány rezistentní a multirezistentní bakteriální kmeny pomocí molekulárně-genetických metod. Výsledky prokázaly, že ARGs se nacházely ve všech studovaných nádržích ČOV a také ve vodním recipientu, do kterého byly vody z ČOV odváděny. Zajímavým zjištěním bylo, že sedimentační fáze čištění ČOV byla ideálním prostředím pro výskyt ampicilin-rezistentních bakterií a pro kumulaci ARGs, zejména genů blaTEM a tetW. V odpadních vodách byly také detekovány vysoce rizikové geny blaNDM-1 a vanA. Tyto geny patří sice ke vzácně se vyskytujícím genům, ale jsou řazeny k rezistentním genům proti antibiotikům poslední záchrany. Jedná se o první studii potvrzující přítomnost genu blaNDM-1 v českých odpadních vodách. Důležité poznatky vyplynuly také ze sledování bakteriální diverzity v odpadních vodách. Byl zjištěn posun mezi výskytem přirozených a rezistentních bakterií v jednotlivých nádržích ČOV. K nejdominantnějšímu taxonu ampicilin-rezistentních bakterií patřila třída Gammaproteobakterie, která může hrát důležitou roli v akumulaci rezistence na beta-laktamové antibiotika v odpadních vodách. Rezistentní izoláty byly identifikovány a byly u nich potvrzeny geny rezistence. U multirezistentních kmenů byly zjištěny současně se vyskytující geny rezistence na beta-laktamy, karbapenemy a tetracykliny. Mezi multirezistentními kmeny dominoval Aeromonas sp., který představuje určité nebezpečí vzhledem k šíření rezistence mezi různými druhy bakterií v životním prostředí. Mezi identifikované multirezistentní kmeny patřil i oportunní patogen Pseudomonas aeruginosa. Další výzkum byl zaměřen na vliv sub-inhibičních koncentrací antibiotika tetracyklinu na tvorbu biofilmu rezistentními izoláty z odpadních vod. Bylo zjištěno, že koncentrace tetracyklinu od 0.031 do 0.125 ?g/mL měly stimulační účinek nejen na bakteriální růst, ale také na tvorbu bakteriálního biofilmu. Rezistentní izoláty tvořící biofilm jsou rizikovým faktorem pro vznik a šíření ARGs v odpadních vodách a dále do životního prostředí.
