Susceptibility testing of clinical isolates of Clostridioides difficile to selected antibiotics by E-test

Abstract:

Cílem bakalářské práce je stanovit citlivost u klinických izolátů Clostridioides difficile z let 2011 a 2021 na vybraná antibiotika a zhodnocení výsledků naměřené citlivosti k vybraným antibiotikům. Testování citlivosti je provedeno pomocí E-testu. V teoretické části jsou shrnuty poznatky o rodu Clostridium. Je zde popsána bakterie Clostridioides difficile, její výskyt a onemocnění, jehož je původcem. Nachází se zde i diagnostika a léčba onemocnění. V druhé části je probrána historie antibiotik, mechanismy účinku a mechanismy vzniku rezistence na antibiotika, jsou zde uvedeny základní metody testování citlivosti na antibiotika a popis vybraných druhů antibiotik. Praktická část se zabývá popisem získaných izolátů Clostridioides difficile, jejich kultivací a následným stanovením citlivosti pomocí E-testu. Jsou zde uvedena kultivační média a použité přístroje. K testování byla použita antibiotika erytromycin, clindamycin, metronidazol a ciprofloxacin. Dosažené rezistence jsou velmi variabilní a liší se od výsledků studií z příslušných let. Výsledky této práce prokazují důležitost stanovení lokálního profilu rezistence tohoto patogena na konkrétní antibiotikum.

Abstract:

The aim of this bachelor thesis is to determine the sensitivity of clinical isolates of Clostridioides difficile to selected antibiotics and to evaluate the results of the measured sensitivity to selected antibiotics. Sensitivity testing is performed by E-test. The theoretical part summarizes the knowledge about the genus Clostridium. There are described bacterium Clostridioides difficile, its occurrence and the disease it causes. There is also a diagnosis and treatment of the disease. The second part discusses the history of antibiotics, mechanisms of action and mechanisms of antibiotic resistance, there are standard testing methods of antibiotic sensitivity and a description of selected types of antibiotics. The practical part deals with the description of the obtained isolates of Clostridioides difficile, their cultivation and subsequent testing of sensitivity by E-test. The culture media and equipment used are listed here. The antibiotics erythromycin, clindamycin, metronidazole and ciprofloxacin were used for testing. The achieved resistances are very variable and differ from the results of studies from the relevant years. The results of this work demonstrate the importance of testing the local resistance profile of this pathogen to a particular antibiotic.
