Parameters of oxidative stress in selected pathological conditions

Abstract:

Oxidative stress is defined as imbalance between prooxidative and antioxidative factors and up to the present time, the oxidative stress was linked to many diseases and pathological conditions. The aim of the thesis was a determination of two parameters of oxidative stress in different biological materials and in different pathological conditions. These parameters were 8-hydroxy-2’-deoxyguanosine as a parameter of oxidative damage of DNA and 4-hydroxy-2-nonenal as a final product of lipid peroxidation. Beside these two parameters, additional parameters were determined to clarify the problem studied. The first part of the thesis examined the susceptibility of isolated rabbit hearts with increased mass of left ventricle to repeated cycles of ischemia and reperfusion using biochemical and electrophysiological parameters. The second part of the thesis examined the effect of different doxorubicin formulation on selected biochemical parameters and gene expression in rats to assess the severity of doxorubicin-induced oxidative stress. In the third part of the thesis the time course of oxidative stress in severely burned patients was examined. Beside 8-hydroxy-2’-deoxyguanosine and 4-hydroxy-2-nonenal, comet assay was newly used in this condition.

Abstract:

Oxidačný stres je definovaný ako nerovnováha medzi prooxidačnými a antioxidačnými faktormi a do dneška bol spojený so širokou škálou ochorení. Práca je zameraná na stanovenie dvoch parametrov oxidačného stresu v rôznych biologických materiáloch a v rôznych patologických stavoch. Týmito parametrami sú 8-hydroxy-2‘-deoxyguanozín, parameter oxidačného poškodenia DNA, a 4-hydroxy-2-nonenal, jeden z konečných produktov lipoperoxidácie. Okrem nich boli stanovené v každej štúdii ďalšie doplnkové parametre, ktoré pomáhali dokresliť celkový obraz skúmanej problematiky. Prvou štúdiou bol vplyv zväčšenej hmotnosti ľavej komory na citlivosť myokardu k opakovaným cyklom ischémie a reperfúzie pomocou hodnotenia biochemických a elektrofyziologických parametrov. Druhá štúdia hodnotila vplyv rôznych foriem doxorubicínu na vybrané biochemické parametre a expresiu génov u laboratórneho potkana pre posúdenie závažnosti doxorubicínom indukovaného oxidačného stresu. V poslednej štúdii bol sledovaný časový priebeh parametrov oxidačného stresu u stredne ťažko až závažne popálených pacientov v priebehu hospitalizácie. Okrem 8-hydroxy-2‘-deoxyguanozínu a 4-hydroxy-2-nonenalu bola na sledovanie využitá tiež po novom kométová analýza cirkulujúcich leukocytov.
