Polymorphism of genes ABC transporters in relation to colorectal cancer disease

Abstract:

Cílem této práce bylo zjistit potenciální vliv mutace genu CFTR, člena rodiny ABC transportérů, na patogenezi kolorektálního karcinomu. V teoretické části je nejprve popsána patogeneze nádorového onemocnění, další kapitola je věnována kolorektálnímu karcinomu, jeho patogenezi, diagnostice, klinickému průběhu a léčbě. Dále je popsána stavba a funkce ABC transportérů, zvláštní kapitola je věnována genu CFTR a cystické fibróze. Na závěr teoretické části jsou uvedeny nejčastější metody pro detekci mutací a polymorfismů genomové DNA a popis metody ASA PCR použité v praktické části práce. Praktická část začíná definováním cílů a metodikou práce. Dále je popsán postup izolace DNA, PCR a analýzy amplifikovaných produktů elektroforézou. Následují naměřené hodnoty izolované DNA a výsledky stanovení mutace delF508 v genu CFTR u pacientů s kolorektálním karcinomem. Bakalářská práce je zakončena diskuzí metodických problémů a výsledků. Závěr shrnuje dosažené výsledky a dává další perspektivu výzkumu uvedené problematiky.

Abstract:

The purpose of this work was to investigate the potential impact of mutation of the CFTR gene, a member of ABC transporters, on the pathogenesis of colorectal cancer. At the begining of the theoretical part, the pathogenesis of cancer is described. The next chapter deals with colorectal carcinoma, its pathogenesis, diagnosis, clinical course and treatment. Then the ABC transporter's structure and function is described. A specific chapter is dedicated to the CFTR gene and cystic fibrosis. At the end of the theoretical part there are presented the most frequent methods for detection of genomic DNA mutations and polymorphisms and a description of the ASA PCR method used in the practical part of the thesis. The practical part begins with the definition of the objectives and methodology of the work. The procedure of DNA isolation, PCR and analysis of amplified products by electrophoresis is described afterwards. Following are the measured values of isolated DNA and the results of delF508 mutation determination in the CFTR gene of patients with colorectal cancer. The bachelor thesis ends with discussion of methodological problems and results. The conclusion summarizes the results and gives a further perspective to the research.
