Mitochondrial variation of the brown bear in Western Carpathians

Abstract:

Předložená práce měla za cíl vytvořit literární přehled týkající se mitochondriální variability medvěda hnědého (Ursus arctos) na světě a diskutovat zjištěné informace s výsledky získanými v praktické části. Sběr vzorků u obou populací ze Západních i Východních Karpat na Slovensku byl zajištěn Štátnou ochranou prírody Slovenskej republiky (ŠOPSR). K analýze vzorků byla použita kontrolní oblast mitochondriální DNA (mtDNA). Výsledky ukázaly výskyt dvou odlišných mitochondriálních linií na území Slovenska. Haplotyp náležící k linii 1b byl nalezen pouze na východě Slovenska, které geomorfologicky spadá pod Východní Karpaty. Haplotypy linie 3a byly nalezeny především ve střední části Slovenska, spadající pod Západní Karpaty, avšak jeden vzorek byl nalezen i ve Východních Karpatech. Tento výsledek může svědčit buď o recentním propojení obou populací po jejich rozdělení během první světové války, nebo je tato fylogeografie dána procesy, které se udály například již během období LGM (last glacial maximum) a následné rekolonizace střední Evropy.

Abstract:

The aim of this thesis was to create a literary overview of the mitochondrial variability of brown bears (Ursus arctos) in the world and then discuss the findings with the results obtained in the practical part. Collection of samples from both Western and Eastern Carpathians populations in Slovakia was realized by the State Nature Conservation of the Slovak Republic. Mitochondrial DNA control region was used to analyze the samples. The results showed the occurrence of two distinct mitochondrial lineages in the Slovakia. The haplotype from the lineage 1b was found only in the east of Slovakia, which geomorphologically falls to the Eastern Carpathians. The haplotypes of lineage 3a were found mainly in the central part of Slovakia, situated in the Western Carpathians, but one sample of this lineage was found also in the Eastern Carpathians. This result may indicate, firstly, the recent connecting of the two populations after their separation during the First World War. The second option is that this phylogeography is caused by the processes that took place during the LGM (last glacial maximum) and during the later recolonization of Central Europe.
