Preparation of ligands for novel types of metallodrugs

Abstract:

Tato bakalářská práce se v teoretické části zabývá cyklodextriny, jejich farmaceutickými aplikacemi a možnou interakcí cyklodextrinu s adamantanem. Molekula adamantanu byla v naší práci vybrána z důvodu vysoké afinity ke kavitě cyklodextrinu. Příprava a použití ligandů nesoucích adamantyl při přípravě metaloléčiv by mělo napomoci vložení molekuly metaloléčiva do ochranné cyklodextrinové kapsule cílící na rakovinné buňky. Dále se věnuji jednotlivým druhům metaloléčiv a poté také dvěma druhům cross-couplingů, Suzukimu couplingu a Negishimu couplingu. Tyto dvě reakce jsme vybrali jako možné cesty přípravy ligandů odvozených od adamantanu, 3- a 4-adamantylpyridinu, které by byly schopny interagovat jak s cyklodextrinem, tak vytvářet koordinační vazbu s biologicky aktivním kationtem kovu prostřednictvím pyridinu. Praktická část poté navazuje přípravou 3- a 4-adamantylpyridinu, a to jak pomocí Suzukiho, tak Negishiho couplingu. Optimalizací reakčních podmínek pak konečně získáváme v nízkých výtěžcích ligand 4-adamantylpyridinu. Další optimalizace výtěžnosti reakcí budou následovat.

Abstract:

This thesis deals with cyclodextrins, their pharmaceutical applications, and possible interactions with adamantane. Adamantane moiety has been chosen because of its high affinity towards cyclodextrin cavity. Preparation and use of ligands bearing adamantyl moiety in preparation of metallodrugs should enable incorporation of metallodrug into protective and cancer-targeting cyclodextrin capsule. Furthermore, it describes different kinds of metallodrugs and two cross-coupling reactions – Suzuki coupling and Negishi coupling. Those are the two possible ways for preparation of ligands derived from adamantane, 3- and 4-adamantylpyridine, which can interact with both, cyclodextrin and also form coordination bond with biologically active metal cation. The experimental part then deals with the preparation of 3- and 4-adamantylpyridine via both Suzuki and Negishi coupling and shows which reaction is more suitable. After optimization of reaction conditions, we finally obtained 4-adamantylpyridine ligand in low yields. Further optimizations of the reactions are ongoing.
