Methanogens in the digestive tract of gorillas

Abstract:

Tato práce je zaměřena na zjištění výskytu metanogenních archeí v zažívacím traktu goril nížinných a horských. V první části je pojednáno obecně o gorilách, jejich výskytu, morfologii, stravě v přirozeném prostředí a při chovu v zoologických zahradách, a o složení a mikrobiálním osídlení jejich zažívacího traktu. První část také pojednává o metanogenních archeích, jejich systematice a vlastnostech a základních molekulárně genetických metodách. Druhá část práce je zaměřena na metodiku výzkumu výskytu metanogenů v trávicí soustavě goril nížinných a goril horských vybranými metodami molekulární biologie. Cílem práce bylo zjistit, zda se v trávicím traktu goril nížinných a horských žijících ve volné přírodě vyskytují metanogenní archea a jaké je jejich taxonomické zařazení. Celkem bylo analyzováno 14 vzorků DNA izolovaných z trusu goril nížinných a goril horských, ale jen 6 vzorků bylo dále analyzováno. Sekvenační analýzou bylo prokázáno, že detekované metanogeny jsou blízce příbuzné kultivovaným druhům Methanobrevibacter smithi. Ostatní detekované metanogeny reprezentují nekultivované klony blízce příbuzné klonům vyskytujícím se v zažívacím traktu orangutana, krávy, prasete, yaka a také člověka. Vzorky byly zkoumány pomocí molekulárně-genetických postupů, jako jsou denaturační gradientová gelová elektroforéza (DGGE) a polymerázová řetězová reakce (PCR). Pomocí sekvenační analýzy 16S rDNA byli identifikovány metanogeny v zažívacím traktu goril.

Abstract:

This work is focused on detecting the presence of methanogenic archaea in the digestive tract of lowland and mountain gorillas. The first part deals generally about gorillas, their occurrence, morphology, diet and the natural environment for breeding in zoos, and the composition and microbial colonization of the digestive tract. The first part also discusses the methanogenic Archaea, their scheme and basic properties and molecular genetic methods. The second part focuses on the research methodology incidence of methanogens in the digestive tract of lowland gorillas and mountain gorillas selected methods of molecular biology. The aim of the study was to determine whether the digestive tract of lowland and mountain gorillas living in the wild methanogenic archaea and what is their taxonomic classification. A total of 14 samples were analyzed DNA isolated from the feces of lowland gorillas and mountain gorillas, but only 6 samples were further analyzed. Sequencing analysis showed that the detected methanogens are closely related to the cultivated species Methanobrevibacter smithii. Other detected methanogens represent wild-type clones closely related clones occurring in the digestive tract, orangutan, cow, pig, yak and humans. Samples were examined using molecular genetic techniques, such as denaturing gradient gel electrophoresis (DGGE) and polymerase chain reaction (PCR). Using sequencing analysis of the 16S rDNA were identified methanogens in the digestive tract gorillas.
