Evaluating acute toxicity of sediments from the Pocheň water reservoir using tests for Eisenia foetida and Lactuca sativa

Abstract:

Bakalářská práce se zabývá problematikou znečištění dnových sedimentů vodné nádrže Pocheň, jež byly z nádrže v letech 2020-2021 odtěženy. Teoretická část práce je zaměřena na popis vzniku sedimentů a jejich struktury. Dále je přiblížena funkce sedimentů a negativní vlivy působící na sedimentaci a je rozvedena problematika kontaminace vody a dnových sedimentů. V teoretické části práce jsou popsány také principy ekotoxikologie a ekotoxikologické testy užívané k testování dnových sedimentů. Experimentální část práce hodnotí akutní toxicitu vytěžených sedimentů z vodní nádrže Pocheň za použití testů akutní toxicity na žížalách (Eisenia foetida) a na salátu (Lactuca sativa). V experimentální části práce jsou charakterizovány testovací organismy, standardní látky vybrané pro ověření správnosti provedení pokusu, postup odběru a příprava vzorků před testem. Dále jsou popsány metodiky použité k provedení testů na standardních látkách i na reálných vzorcích sedimentů, včetně interpretace naměřených dat.

Abstract:

The bachelor's thesis deals with the issue of pollution of the bottom sediments of Pocheň water reservoir, which were extracted from the reservoir in 2020-2021. Theoretical part of the thesis is focused on the description of the formation of sediments and their structure. Furthermore, the function of sediments and the negative effects on sedimentation are approached and the issue of contamination of water and bottom sediments is discussed. Theoretical part of the thesis also describes the principles of ecotoxicology and ecotoxicological tests used to test bottom sediments. Experimental part of the work evaluates the acute toxicity of extracted sediments from the Pocheň reservoir using acute toxicity tests on earthworms (Eisenia foetida) and lettuce (Lactuca sativa). The experimental part of the work characterizes testing organisms, standard substances selected to verify the correctness of the experiment, sampling and preparation of samples before the test. The methodologies used to perform tests on standard substances and on real sediment samples are also described, including the interpretation of measured data.
