Supramolecular Host Molecules for Anion Binding

Abstract:

V této disertační práci se zabýváme přípravou neutrálních receptorů aniontů a studiem jejich supramolekulárních vlastností. V teoretické části jsou popsány neutrální receptory aniontů se zvláštním přihlédnutím k receptorům, které váží anionty ve vodném prostředí. Experimentální část práce je složena ze tří projektů. První projekt popisuje syntézu dimerních klips propandimočoviny a zkoumá jejich supramolekulární vlastnosti. Bylo zjištěno, že tyto klipsy jsou schopné vázat anionty a vytvářet tak komplexy se stechiometrií 1:1. Pozorované komplexy propandimočoviny s anionty by mohly pomoci vysvětlit úlohu aniontů při syntéze cucurbiturilů. V druhém projektu byl navržen a připraven dimerní receptor aniontů založený na monofunkcionalizovaném biotinsulfon[6]urilu. V kapitole je popsána charakterizace tohoto receptoru pomocí NMR a MS a také jeho supramolekulární vlastnosti, které byly zkoumány pomocí ITC. V poslední části disertace je popsán nový typ makrocyklu složený ze stavebních jednotek 2,4 dimethylglykolurilu and m-xylylenu. Byly izolovány jeho dvou- a tří- členné homology. Dvoučlenné makrocykly se vyskytovaly ve dvou diastereomerních formách, které mohly být, díky flexibilitě glykolurilových jednotek, přítomny ve dvou konformacích. Tyto makrocykly byly charakterizovány pomocí NMR a MS a jejich konformační vlastnosti byly zkoumány pomocí VT NMR. Pomocí MS bylo také pozorováno, že tříčlenné makrocykly tvoří adukty s anionty.

Abstract:

In this thesis we present the synthesis of non-charged anion receptors and investigation of its supramolecular properties. The theoretical part describes neutral anion receptors with emphasis on those which bind anions in aqueous environment. The experimental part of the thesis consists of three projects. The first project describes the synthesis of xylylene-walled propanediurea dimers and investigation of its supramolecular properties. It was found that these clips are able to bind anions with 1:1 soichiometry. The observed complexes of propanediurea dimers with anions could help to clarify the role of anions during cucurbiturils formation. In the second project, dimeric anion receptor was designed and synthetized using monofunctionalized biotinsulfone[6]uril macrocycle. The chapter shows characterization of the dimeric receptor by NMR and HRMS, followed by a study of its supramolecular properties by ITC. The last part of this thesis describes a new type of hybrid macrocycle consisting of 2,4 dimethylglycoluril and m-xylylene building blocks. Two and three membered homologues of such macrocycle were isolated. Two diastereomers of two membered macrocycles were isolated, each isomer occurs in two conformations due to the flexibility of glycoluril unit. The two membered macrocycles were characterized, and the properties of the conformers were further studied by methods of VT-NMR. The three membered macrocycles were observed by MS to form adducts with anions.
