Digestion of bryophytes tissues by groundhoppers of genus Tetrix

Abstract:

Diplomová práce je zaměřená na trávení pletiv mechorostů maršemi rodu Tetrix. Jedním z cílů této práce bylo testování metody barvení obsahu buněk mechorostů pro účely porovnávání rozdílů v míře natrávení fragmentů mechů ze střeva a z trusu. Testováno bylo 7 barviv, ale žádné z nich neobarvilo mech v požadované míře. Dalším cílem bylo studium potravní biologie pomocí dvou různých metod. První z nich byla metoda výběru spektra zelené barvy chloroplastů. Vyhodnoceno bylo celkem 200 snímků ze vzorků střeva a trusu 10 jedinců marší Tetrix tenuicornis. Bylo zjištěno, že zastoupení zelené barvy na ploše fragmentu se neliší mezi vzorky z trusu a ze střeva. Druhá metoda spočívala v měření fragmentů mechu a následného porovnání. Analýza ukázala, že fragmenty ve střevě jsou menší, než fragmenty v trusu, což je zřejmě důsledkem nepřesnosti použité metody. Výsledky ukazují na nízkou míru trávení mechorostů, marše nevyužívají vnitřní obsah buněk natolik, aby se změnilo zastoupení chloroplastů.

Abstract:

The thesis focuses on digesting moss?s tissues by groundhoppers of genus Tetrix. One of the goals of the thesis is to test the method of dyeing moss?s cell?s contents in order to compare and contrast the differences in the extent of digesting moss fragments from intestines and droppings. Seven dyes were tested but none of them dyed the moss in the desired degree. Another goal of the thesis is to explore the food biology using two different methods. The first one is the method of choosing the spectrum of chloroplast green colour. 200 photographs of 10 Tetrix tenuicornis groundhopperses intestines and droppings were assessed. The representation of the green colour on the fragment?s surface was found to be the same both in the droppings and in the intestines. The second method is based on measuring the moss fragments and a consequent comparison. The analysis showed that the fragments in the intestines were smaller than the fragments in the droppings. However, such a result is impossible. The inaccuracy may lie in a different amount of food which the preparations had been made from. The results show a low extent of digesting moss. Groundhoppers do not use the inner cell?s content enough for the representation of chloroplasts to be changed.
