Molecular, cellular, and morphological aspects of odontogenesis in selected model species

Abstract:

Zuby, jakožto jeden z klíčových orgánů pro získávání a zpracování potravy u řady živočichů, se dostávají čím dál více do popředí zájmu nejen lékařů, ale i vědců. Kromě čistě klinických studií, zaměřujících se na odontologii a aplikaci nových léčebných metod, jsou zuby často skloňovány též v kontextu odontogeneze a aspektů s ní souvisejících. Poznatky z těchto studií přispívají nejen k hlubšímu pochopení procesů zásadních pro správný vývoj chrupu, ale lze je rovněž vztáhnout na široké spektrum dalších vědních disciplín.Jeden z recentních směrů výzkumu odontogeneze je zaměřen na evoluční aspekty vývoje zubů jako je jejich heterogenita v tvaru, počtu zubních generací a ve způsobu upevnění zubů v kosti čelisti. Často se pro studium tohoto fenoménu využívají plazí modely, jejichž anatomická a morfologická specifika společně s možnostmi analýz představujeme v rámci jedné z kapitol této dizertační práce. Rovněž jsme se zaměřili na mechanizmy, které se podílejí na určování finálního tvaru plazích zubů. Zjistili jsme, že plazi disponují strukturou podobnou savčímu sklovinnému uzlu, která v raných stádiích vývoje slouží jako signalizační centrum řídící iniciální fáze tvarování zubních zárodků. V pokročilejších stádiích vývoje je pak finální tvar zubu dán změnami v uspořádání buněk, způsobenými mimo jiné reorganizací cytoskeletu. Sledován byl i původ a vývoj vaječného zubu jakožto unikátní struktury typické právě pro zástupce plazů, který dle našich zjištění vzniká ze dvou oddělených zubních základů, splývajících následně v jeden celek. Zároveň bylo analyzováno, jakým způsobem je zajištěno pevné připojení zubu ke kosti čelisti, zvané akrodontní ankylóza., V případě výskytu ankylózy u savců se jedná o patologický stav, vznikající jako reakce na porušení měkkých tkání obklopujících zub v zubním alveolu. V této souvislosti byla popsána nová, přechodně se vyskytující populace buněk nazvaných ankyloblasty, které doposud nebyly známy. Pro lepší pochopení fyziologické i patologické ankylózy byly použity vzorky jak původem z plazích druhů, tak vzorky ze zubů extrahovaných pacientům trpícím ankylózou. Za účelem bližšího pochopení patogeneze byly tyto zuby podrobeny analýze s cílem určit rozhraní tkání zubu a kosti za pomocí metody LIBS. V neposlední řadě byla v rámci zpracování této dizertační práce věnována pozornost procesům na buněčné a molekulární úrovni. Blíže byl popsán zejména fenomén zapojení apoptózy do odontogeneze. Apoptóza se zdá být univerzální součástí vývojových procesů organizmů sloužící nejen k eliminaci cílových buněk, ale rovněž i k morfogenezi tkání a orgánů. Kromě buněčné smrti byl sledován také fenomén polarizace buněk náhradní zubní lišty, a to u prasat s difyodontní denticí. Tato struktura nesoucí buňky, které dávají vzniknout náhradní zubní generaci, se v průběhu vývoje zanořuje hlouběji do mezenchymu a její růst vykazuje prostorovou asymetrii provázenou i asymetrickou expresí proteinů spojených s WNT signalizací. Závěrem lze konstatovat, že problematika odontogeneze je velmi obsáhlá a v rámci tohoto tématu lze studovat celou řadu aspektů. V předkládané dizertační práci byly některé z nich poodhaleny, u jiných naopak vyvstala řada dalších otázek, které na své zodpovězení teprve čekají.

Abstract:

Teeth, as one the key organs for uptake and food processing in most of living animals, are recently in the spotlight of the scientists as well as the clinicians. Besides studies focused on new surgical and treatment procedures, also the basic knowledge about how teeth develop during the odontogenesis and what are the key mechanisms behind this process, are needed. All knowledge coming from the recent studies, helps us to understand regulatory processes, which are necessary for the proper teeth development and dental tissue reparation, however can be potentially useful for understanding other scientific questions related to epithelial-mesenchymal interactions during morphogenesis. One of recent tooth-related research focuses on evo-devo aspects concentrating on developmental processes leading to large heterogeneity of tooth shape, number of tooth generations or morphology of tooth-bone attachment. Reptilian species are often used as model organisms to follow these topics from comparative point of view. Therefore, in our research, we selected several representatives from Reptilia class to demonstrate diverse anatomical and morphological features of dentition. Also, we focused on the question, which cellular mechanism is responsible for such heterogeneity especially for different tooth shape. In reptiles, we revealed cell cluster, noticeably similar to mammalian enamel knot, which is known to drive early tooth morphogenesis. During later mineralisation stages, the shape of reptilian tooth crown is determined by the cell shape shift partially caused by cytoskeleton reorganization. We also followed the origin of egg tooth, another structure uniquely found in reptiles. Egg tooth was found to be arising from two separated tooth germs, which gradually merged in one entity. Another interesting feature is physiologically occurring acrodont ankylosis represented by the firm attachment of tooth and bone in some of reptile species. In mammals, ankylosis is initiated when soft tissues surrounding alveolus is damaged by trauma, which is in contrast to reptilian species where ankylosis occurs early in development. In chameleon, we described new transient cell population located at the tooth-bone interface in chameleons, which we called ankyloblasts. These cells are responsible for the production of interface tissue and so far, their presence have not been described yet. For better understanding of processes leading to pathological ankylosis in human, we also evaluated the teeth from the human patients suffering by this clinical manifestation. While using LIBS, we were able to assess the tooth-bone interface and the differences in mineral composition between the bone/tooth and interface tissue mediating the ankylosis. Finally, we also paid the attention to developmental processes during odontogenesis on molecular and cellular level. Apoptosis was found to be involved in odontogenesis and seems to play an important role not just in elimination of cells in specific localized areas but also is crucial for tooth shaping. Moreover, cell polarity was studied in context of successional dental lamina growth and maintenance. This structure plays a crucial role in the initiation of new tooth generations in mammals as well as reptiles. Angled growth of this structure into the mesenchyme seems to be associated on asymmetrical expression of proteins related to WNT signalling. In summary, odontogenesis offer a great potential to study multiple developmental topics from different aspects. In this thesis, we focused on some of these perspectives, however many questions still remain unanswered and thus waiting to be clarified.
