Study of the mechanism of photochemical reactions using time-resolved spectroscopic methods

Abstract:

Ph.D. projekt byl zaměřen na studium mechanismů fotochemických a fotofyzikálních procesů. Získané výsledky shrnuty v této disertační práci jsou již publikovány nebo jsou sepsány a připraveny k publikaci. V teoretické části práce jsou shrnuty základní informace příslušných témat vztahujících se k studovaným projektům, jako jsou fotofyzikální a fotochemické procesy, transientní spektroskopie, xanthenové chromofory a jejich využití při biologických studiích a jsou přestaveny použité sloučeniny - bambusurily a methylová violeť. V následující části práce jsou diskutovány výsledky tří samostatných projektů. Nejprve je zmíněna komplexní studie kinetiky a optických vlastností transientů bengálské červeně vzniklých po excitaci zeleným světlem nanosekundovou transientní spektroskopií. Tato detailní analýza byla dále použita v následujícím projektu zabývajícím se dimerizací N-acetyl tyrosinu a tryptofanu fotosenzitizací bengálskou červení ve vodném fosfátovém pufru se zaměřením na mechanismus této reakce. Bylo zjištěno, že dimerizace nastává v anoxických podmínkách, kde bengálská červeň slouží jako oxidant, který sám podléhá degradaci, zatímco v přítomnosti kyslíku tento proces je v kompetici s účinnou oxidací obou aminokyselin singletovým kyslíkem senzitizovaným bengálskou červení. Jako poslední je uveden studovaný fotoindukovaný přenos elektronu v krystalech hexafluorofosforečnanu methylové violeti a makrocyklu dodekamethylbambus[6]urilu. Původ vzniklého dlouze žijícího iontového radikálového páru v tomto fotoaktivním krystalickém materiálu byl studován a vysvětlen pomocí experimentálních a výpočetních postupů.

Abstract:

The Ph.D. studies were focused on the mechanistic investigation of photochemical and photophysical processes. The obtained results summarized in this thesis have either been published or will be submitted for publication. In theoretical part, basic information about the relevant topics involved in the studied projects, such as photophysical and photochemical processes, transient spectroscopy, xanthene chromophores and their main biological applications, and other chemical moieties used in this work, bambusurils and methyl viologen, are introduced. In the next part, results of three separate projects are discussed. First, the comprehensive investigation of the complex mixed-order kinetics and the optical properties of rose bengal species formed upon visible light excitation was performed by nanosecond transient absorption spectroscopy. This detailed analysis was then used in the subsequent project dealing with crosslinking of N-acetyl tyrosine and tryptophan by photosensitization using rose bengal in phosphate buffer saline solution and with the mechanism of this reaction. It was found that crosslinking takes place in anoxic conditions with rose bengal as a sacrificial oxidant, whereas this process competes with the efficient oxidation of both amino acids by singlet oxygen produced by rose bengal sensitization in aerated solutions. Finally, the photoinduced electron transfer process in co-crystals of methyl viologen hexafluorophosphates and the macrocycle dodecamethylbambus[6] uril was investigated. The origin of the persistent charge-separated state in this photoactive crystalline material was examined and explained by a combined experimental–computational approach.
