Isolation and typing of bacteria resistant to critically important antibiotics in food-producing animals

Abstract:

Zatímco v humánní medicíně zaujímá kolistin ojedinělé postavení při léčbě život ohrožujících infekcí, ve veterinární sféře je hojně aplikován k profylaxi a terapii běž-ně se vyskytujících bakteriálních infekcí způsobených neinvazivními kmeny čeledi Enterobacteriaceae, zejména E. coli. Plazmidy zprostředkovaná rezistence ke kolisti-nu kódovaná geny mcr byla poprvé zdokumentována v roce 2015, a dodnes byla de-tekována v různých zdrojích po celém světě s významným zastoupením mezi hospo-dářskými zvířaty. Cílem práce bylo zjistit výskyt E. coli s rezistencí ke kolistinu ve vzorcích potravinových zvířat a charakterizovat izoláty s geny mcr. Studií jsme prokázali vysoký záchyt rezistentních izolátů E. coli nesoucích gen mcr-1 v čerstvém drůbežím mase, a to zejména v mase z dovozu; naopak ve vzorcích sle-pých střev jatečních zvířat byl výskyt minimální. Fenotypovými testy rezistence a vyhodnocením dat z celogenomového sekvenování byla u těchto izolátů zjištěna vy-soká míra rezistence k dalším antibiotikům, přičemž všechny izoláty vykazovaly mul-tirezistenci k třem a více antibiotikům. Konjugačním experimentem byla zjištěna přítomnost genu mcr-1 na epidemických plazmidech, které se mohou šířit mezi bak-teriemi napříč populacemi. Práce přinesla nové poznatky o epidemiologii kolistinové rezistence v potravinách a u hospodářských zvířat pocházejících z České republiky a přispěla k objasnění me-chanismu šíření genů mcr v čeledi Enterobacteriaceae.

Abstract:

While in human medicine colistin holds a unique position in the treatment of life-threatening infections, in the veterinary sphere it is widely used for prophylaxis and therapy of common bacterial infections caused by non-invasive strains of the Entero-bacteriaceae family, especially E. coli. Plasmid-mediated colistin resistance encoded by the mcr genes was first documented in 2015, and to date has been detected in var-ious sources worldwide with a significant prevalence among livestock. The aim of this study was to determine the occurrence of colistin-resistant E. coli in samples of food animals and to characterize isolates with mcr genes. The results showed a high occurrence of resistant E. coli isolates carrying the mcr-1 gene in fresh poultry meat, especially in imported meat; in contrast, the prevalence in caecal samples from slaughter animals was low. Phenotypic resistance testing and whole-genome sequencing data of these isolates revealed high levels of resistance to other antibiotics, with all isolates showing multidrug resistance to three or more an-tibiotics. Conjugation experiment showed the presence of the mcr-1 gene on epidemic plas-mids that can spread between bacteria across populations. The study has provided new insights into the epidemiology of colistin resistance in food and livestock originating from the Czech Republic and contributed to the eluci-dation of the mechanism of mcr genes spread in the Enterobacteriaceae family.
