The capacity of pythin oligandrin induce resistence in tomato plants

Abstract:

Imunitní reakci u rostlin spojenou s rezistencí vůči patogenům vyvolávají látky, které souhrnně nazýváme elicitory. Mezi elicitory patří také malé proteiny elicitiny, které jsou produkované oomycetami rodu Phytophthora a Pythium. Jedním z elicitinů je oligandrin, protein o velikosti okolo 10 kDa, který byl izolovaný z oomycety Pythium oligandrum. Tato bakalářská práce se věnuje schopnosti elicitinu oligandrinu indukovat širokospektrální rezistenci u modelové rostliny rajčete Solanum lycopersicum cv. Micro-Tom. Nejdříve byl zjišťován nejvhodnější způsob aplikace proteinu oligandrinu do rostlin rajčete a to buď pomocí nasávání roztoku oligandrinu přes listovou petiolou, nebo přímým nástřikem do apoplastu listů. Přímá infiltrace způsobovala nežádoucí nekrózu listu, proto byla dále používána pouze aplikace pomocí nasávání roztoku oligandrinu listovou petiolou. Schopnost oligandrinu indukovat imunitní odpověď byla následně určována prostřednictvím míry rezistence rostlin rajčete (Solanum lycopersicum cv. Micro-Tom) proti bakterii Pseudomonas syringae pv. tomato. Ke stanovení míry rezistence byla použita real-time PCR, kde byla jako DNA templát použita DNA izolovaná z listů s aplikovaným oligandrinem (1 a 5 µM) a P. syringae pv. tomato. Byla provedena kvantifikace genu pro conserved domain protein (lokus PSPTO_4345) P. syringae a genu Tip41 rajčete. Následně byly tyto výsledky ověřeny kultivací bakterie P. syringae pv. tomato z listů s oligandrinem (5 µM) na King´s B mediu. Bylo zjištěno, že 1 µM oligandrin významně neovlivňuje rezistenci, ale 5 µM oligandrin už vyvolává signifikantní zvýšení rezistence.

Abstract:

Plant immunity connected with resistance against pathogens is induced by substances called elicitors. Elicitors include small proteins elicitins secreted by the oomycetes Phytophthora and Pythium. One of the elicitins is oligandrin, a 10 kDa protein isolated from the oomycete Pythium oligandrum. In this thesis, we study the capacity of pythin oligandrin to induce wide-spectrum resistance in model tomato plants Solanum lycopersicum cv. Micro-Tom. We first explored the best method of applying oligandrin on tomato plants either by using absorption of the oligandrin solution by leaf petiole or by direct infiltration. Direct infiltration caused unexpected leaf necrosis. Therefore, we used only the application by an absorption of the oligandrin solution by the leaf petiole. The capacity of pythin oligandrin to induce immune response was determined by the rate of resistance in tomato plants (Solanum lycopersicum cv. Micro-Tom) against bacteria Pseudomonas syringae pv. tomato. The measure of resistance was monitored using real-time PCR with DNA isolated from leaves with applied oligandrin (1 a 5 µM) and P. syringae pv. tomato as a template. A quantification of the gene for conserved domain protein (locus PSPTO_4345) from P. syringae and gene Tip41 from tomato was done. The results were subsequently verified by the cultivation of the bacteria P. syringae pv. tomato from the leaves with applied oligandrin (5 µM) on King´s B medium. While 1 μM oligandrin was found to have no significant effect on resistance, 5 μM oligandrin induced significant resistance.
