The effects of hypoxia on cell differentiation induced with all-trans retinoic acid and LOX/COX inhibitors in osteosarcoma cells

Abstract:

Tumor cells are typically known for their inability to differentiate into mature phenotypes. A high proliferation potential is often connected with this defect, resulting in deregulated growth of tumor cell mass. Differentiation therapy focused on transformed cells differentiation is the most promising approach in this case and often leads to better outcome compared to conventional chemotherapy. Retinoids, especially all-trans retinoic acid (ATRA) are often used for this purpose. It´s instability and substantial toxicity can be overcome with usage of enzyme inhibitors, especially 5-lipoxygenase and cyclooxygenase-2, which slow down catabolism of ATRA. A big attention in the field of cancer therapy is currently focused on the tumor microenvironment and its changes due to the deregulated proliferation of tumor cells. Namely, hypoxia – reduced tissue oxygen concentration, which may cause therapy resistance and lead to worse patient´s prognosis. The goal of diploma thesis is to determine the impact of hypoxia on ATRA´s ability to induce tumor cells differentiation. For this purpose, Saos-2 osteosarcoma cell line was chosen. Changes in proliferation, cell cycle and differentiation were evaluated in treated cell populations. Analysis of hypoxia impact on ATRA and LOX/COX inhibitors ability to reduce proliferation showed negative effect of hypoxia on these mechanisms. However, further analysis of cell cycle suggested that this change of cell quantity depending on used cultivation conditions is primary caused by mechanism of chemicals´ cytotoxicity rather than their antiproliferation effect. Analysis of hypoxia impact on differentiation effect of used chemicals showed slightly negative influence of hypoxic conditions on expression of differentiation markers that were analyzed with RT-PCR for this purpose. In general, results of this diploma thesis show that hypoxia has a negative impact on antineoplastic effect of ATRA and LOX/COX inhibitors in osteosarcoma cells. Extent of this impact depended on the type of used chemicals as well as on the cultivation period.

Abstract:

Jedním z typických rysů nádorových buněk je porucha schopnosti diferencovat do terminálních fenotypů charakteristických pro danou tkáň. S touto poruchou je spojený vysoký proliferační potenciál, jehož následkem je nekontrolovatelný nárůst masy nádorově transformovaných buněk. Slibným terapeutickým přístupem je v těchto případech diferenciační terapie zaměřená na indukci diferenciace nádorových buněk. Jako induktory diferenciace se využívají retinoidy, hlavně kyselina all-trans retinová (ATRA). Její rychlé odbourávání ze tkání a zvýšenou toxicitu je možné modulovat kombinovaným podáváním s inhibitory lipoxygenáz (LOX) a cyklooxygenáz (COX) účastnících se její katabolismu. V současnosti je v rámci nádorové terapie věnována velká pozornost změnám mikroprostředí způsobeným nadměrnou proliferací transformovaných buněk. Konkrétně se jedná například o hypoxii – sníženou koncentraci kyslíku v tkáni. Ta má za následek častou rezistenci k aplikované terapii a zhoršuje prognózu pacientů. Cílem diplomové práce bylo zjistit, jaký vliv má hypoxie na schopnost kyseliny all-trans retinové a jejích modulátorů indukovat diferenciaci nádorových buněk osteosarkomové buněčné linie Saos-2. Sledovány byly změny proliferace, buněčného cyklu a diferenciace. V případě analýzy vlivu kultivačních podmínek na efektivitu tlumení proliferace ATRA a inhibitorů LOX/COX bylo sice pozorované tlumení jejich účinku v hypoxických podmínkách, po detailnější analýze buněčného cyklu průtokovou cytometrií se však zdá, že příčinou sledovaných změn je pravděpodobně vliv hypoxie na jejich cytotoxické a ne na antiproliferační účinky. Analýza vlivu hypoxie na schopnost činidel indukovat diferenciaci prokázala lehce negativní dopad hypoxických kultivačních podmínek na expresi diferenciačních markerů, které byly za tímto účelem sledovanány pomocí RT-PCR. Celkově je tedy možno získané výsledky interpretovat tak, že hypoxie má negativní vliv na antineoplastický účinek ATRA a inhibitorů LOX/COX. Míra tohoto vlivu závisela na typu použitých činidel a také na době, po kterou byly buňky v daných podmínkách kultivované.
