Enzymatic determination of methanol

Abstract:

Práce se zabývá methanolem, mechanismem jeho toxicity a způsobech stanovení. V teoretické části jsou popsány fyzikální a chemické vlastnosti methanolu, jeho výroba, výskyt v přírodě a povolený obsah v alkoholických nápojích podle evropské legislativy. Je zde pojednáno o pektinu, jako hlavním zdroji methanolu v alkoholických nápojích a jeho hydrolýze pomocí pektinmethylesterasy. Pozornost je věnována i toxikologii methanolu a terapii při intoxikaci a metodám stanovení methanolu, jak pomocí technik analytických, tak enzymatických. V experimentální části práce bylo potvrzeno, že přibližně až tisícinásobný molární nadbytek ethanolu, jako kompetitivní inhibitor, způsobuje výraznou inhibici reakce methanolu s alkoholoxidasou a tudíž i snížení fluorescence. Popsaným způsobem lze tedy methanol snadno detegovat.

Abstract:

This thesis deals with methanol, mechanism of toxicity and methods of determination. The theoretical part describes the physical and chemical properties of methanol, its production, natural occurrence and content in alcoholic beverages by European law. It dealt with the pectin as the main source of methanol in alcoholic beverages and its hydrolysis using pectin methylesterase. Attention is also paid to the toxicology of methanol intoxication, therapy and methods used for determination of methanol, using both analytical and enzymatic techniques. In the experimental part it was confirmed that about a thousand times molar excess of ethanol as a competitive inhibitor causes considerable inhibition of the reaction of methanol with alcohol oxidase and decrease in fluorescence. The described method allows for easy detection of methanol.
