In-vitro study of soybean and red clover isofavones in ruminal fluid

Abstract:

Isoflavony patří mezi sekundární metabolity čeledě bobovitých (Fabaceae), které jsou produkovány rostlinami jako obrana proti stresu. V živočišných organismech fungují jako tzv. fytoestrogeny, které napodobují hormon 17-β-estradiol. Nicméně nejvyšší aktivitu vykazuje equol, který vzniká z daidzeinu a formononetinu činností bakterií např. v rumenu přežvýkavců. Cílem praktické části diplomové práce bylo stanovení kinetiky odbourávání sójových isoflavonů a nárůst equolu v ruminální tekutině v závislosti na přidaném množství 40% sójového extraktu. Nejrychlejší nárůst equolu o 0,59 µg.ml-1.hod-1 byl pozorován během posledních 12 hod u vzorků s přídavkem 25 mg 40% extraktu. Dále byly určeny hladiny jetelových isoflavonů a equolu v ruminální tekutině kultivované v bioreaktoru po dobu 10 dní. Ustalování hladin bylo pozorováno od 6. dne, kdy se koncentrace biochaninu A pohybovaly okolo 0,50 µg.ml-1; genisteinu okolo 0,01 µg.ml 1; formononetinu okolo 0,42 µg.ml-1; daidzeinu okolo 0,04 µg.ml-1 a equolu okolo 28,55 µg.ml-1. Pro detekci koncentrací sójových i jetelových isoflavonů byla použita metoda HPLC na reverzní fázi.

Abstract:

Isoflavones are secondary metabolites of Fabaceae plant family synthesized under stress conditions. These compounds are reffered to as phytoestrogenes, because they are capable to mimic hormone 17-β-estradiol in animals. Nevertheless, the highest activity is connected to equol, which is produced from daidzein and formononetin by bacteria present in cow rumen. The aim of the practical part of thesis was to determine kinetics of degradation of soy isoflavones and kinetics of synthesis of equol in ruminal fluid as a function of added 40% soy extract. The highest speed of equol production, which was 0.59 µg.ml-1 per hour, was observed in samples with 25 mg of added extract during the last 12 hours. Also the levels of red clover isoflavones and equol were defined in ruminal fluid, that was cultivated in bioreactor within 10 days. The concentrations of analytes were stabilized around 0.50 µg.ml-1 for biochanin A; 0.01 µg.ml-1 for genistein; 0.42 µg.ml-1 for formononetin; 0.04 µg.ml-1 for daidzein and 28,55 µg.ml-1 for equol since 6th day. Determination of concentration of both soy and clover isoflavones was carried out by using HPLC on reverse phase.
