Study of microRNAs involved in chemoradiorezistance of glioblastoma multiforme

Abstract:

Úvod a problematika: Multiformní glioblastom (GBM) je nejmalignější nádorové onemocnění mozku s mediánem celkového přežití 12-15 měsíců od stanovení diagnózy. Jedna z možností jak zlepšit prognózu pacientů může spočívat v pochopení příčiny značné chemoradiorezistence GBM buněk. Výsledky současných studií ukazují, že významnou roli v rezistenci GBM k léčbě by mohly hrát mikroRNA (miRNA). Tyto krátké nekódující RNA molekuly posttranskripčně regulují expresi více než 2/3 všech lidských genů, které jsou mimo jiné zapojeny i v regulaci signálních drah asociovaných s rezistencí nádorových buněk k terapii. Metody: Pomocí postupného ozařování třech GBM linií do celkové dávky 60Gy byl in vitro připraven model GBM radiorezistentních buněk. Následně byla jejich radiorezistence ověřena pomocí metody colony formation assay. Rozdíl v expresi miRNA mezi radiorezistentními a kontrolními liniemi byl analyzován pomocí Affymetrix GeneChip miRNA array. Změna expresních hladin vybraných genů a miRNA v buňkách po jejich ozáření byla zhodnocena pomocí RT-qPCR. Výsledky: Výsledky ukázaly významně vyšší radiorezistenci GBM buněčných linií U251-R a T98G-R oproti kontrolním buňkám. Jako více radiorezistentní se jevila rovněž třetí zkoumaná buněčná linie U87MG-R, avšak tato nebyla pomocí použitých metod jednoznačně prokázána. U prvních dvou uvedených radiorezistentních linií došlo po jejich vystavení ionizujícímu záření k rychlému zvýšení exprese vybraných genů zapojených v reparaci DNA. Následně bylo pomocí globální expresní analýzy miRNA odhaleno 114 miRNA se signifikantně rozdílnou expresí u radiorezistentních linií oproti kontrolním liniím. Z těchto rozdílně epxrimovaných miRNA byla k podrobnějšímu zkoumání vybrána miR-31, jejíž hladina ve zkoumaných buněčných liniích v reakci na ionizující záření rychle poklesla. Závěr: V této diplomové práci byly in vitro připraveny radiorezistentní GBM buněčné linie, které při bližším zkoumání vykazovaly vyšší expresní hladiny vybraných reparačních genů v reakci na jejich ozáření. U těchto buněčných linií byly následně odhaleny deregulované hladiny mnoha miRNA, které by mohly hrát významnou roli v radiorezistenci GBM a mohly by být potencionálními cíli k překonání radiorezistence u pacientů postižených tímto onemocněním.

Abstract:

Introduction: GBM is the most malignant brain tumour with the median overall survival of newly diagnosed patients only 12-15 months. One possible option how to improve poor prognosis is to better understand the cause of considerable chemoradioresistance of GBM. Recent studies have been indicating that microRNAs (miRNAs) could play a major role in treatment resistance of GBM patients. These short non-coding RNAs molecules posttranscriptionally regulate expression of more than 2/3 of all human genes which are among other involved in regulation of signalling pathways associated with resistance of tumour cell to therapy. Methods: In vitro model of GBM radioresistant cells was established by fractioned irradiation of three GBM cell lines to total dose of 60Gy. Radioresistance of these cell lines was subsequently validated using the colony formation assay. The difference in the miRNA expression between radioresistant and control cell lines was analysed using Affimetrix GeneChip miRNA arrays. Change in miRNA and gene expression in cells after irradiation was evaluated by RT-qPCR. Results: The results showed significantly higher radioresistance of established U251-R and T98G-R GBM cell lines compared to control cells. Third established radioresistant cell line U87MG-R seemed to be also more radioresistant. However, U87MG-R radioresistance has not been clearly demonstrated by colony formation assay. Irradiation of U251-R and T98G-R led to rapid increase in expression of selected genes involved in DNA repair. Subsequently, using global expression analyses of miRNAs, 114 miRNAs were identified to be significantly differentially expressed between radioresistant and control cell lines. From these 114 deregulated miRNAs, miR-31 was selected for detailed examination. Expression level of this miRNA after irradiation was rapidly decreased in studied GBM cells. Conclusion: In this study, GBM radioresistant cell lines were established in vitro. These cell lines showed higher expression levels of DNA repair genes in response to radiation. Additionally, many miRNA expression levels were identified to be deregulated in radioresistant cells indicating that identified miRNAs could play an important role in radioresistance of GBM and that they could be potential targets to overcome radioresistance of GBM patients.
