Inhibition of growth of freshwater algae by binary mixtures of heavy metals

Abstract:

Tato práce se zabývá akutní toxicitou těžkých kovů a jejich binárních směsí pro sladkovodní zelené řasy Desmodesmus subspicatus. V literární rešerši shrnuje poznatky o ekotoxikologických testech toxicity a o toxicitě směsí. V experimentální části byla zjišťována inhibice růstu Desmodesmus subspicatus solemi tří těžkých kovů (síranu kademnatého, chloridu manganatého a chloridu nikelnatého) a jejich binárními směsmi. Experimentální koncentračně-inhibiční křivky byly proloženy modifikovanou Gompertzovou křivkou, která umožňuje popsat celou závislost dvěma parametry a zároveň vypočítat hodnotu EC50. Tyto parametry byly následně využity pro odhad toxicity směsí pomocí dvou teoretických modelů (modelu nezávislého působení a koncentračně aditivního modelu) Při porovnání inhibičních hodnot získaných z experimentu a hodnot vypočtených pomocí obou modelů bylo zjištěno, že ani jeden z modelů neodpovídá plně experimentálním výsledkům. Lepší odhad výsledků poskytuje koncentračně aditivní model.

Abstract:

This works is focused on the toxicity of heavy metals and their binary mixtures to green fresh water algae Desmodesmus subspicatus. Contemporary knowledge on ecotoxicity testing and toxicology of mixtures is summarized first. Inhibition of growth of Desmodesmus subspicatus by three heavy metal salts (cadmium sulphate, manganese chloride, and nickel chloride) and their binary mixtures was then experimentally obtained. Experimental concentration-inhibition curves for single chemicals were regressed by modified Gompertz curve which enabled parameterization of the dependence as well as calculation of EC50 values. Regression parameters were then used for estimation of mixture toxicity using two theoretical models (independent action model and concentration-addition model). Comparison of experimental and estimated inhibition values showed that none of the models explained experimental data thoroughly. Better estimation was provided by concentration-addition model
