Production of DIRIGENT protein AtDIR14 in Pichia pastoris

Abstract:

Dirigent proteiny v rostlinách řídí regio- a stereoselektivitu fenoxy radikálových kondenzačních reakcí a hrají tak zásadní roli v syntéze biologicky aktivních přírodních produktů, jako jsou lignany či gossypol. Jelikož exprese proteinu AtDIR13 se mění v reakci na cytokinin trans-zeatin a transkripční faktory ARR10 a ARR1, členové cytokininové signální dráhy, mají vysokou afinitu k promotoru AtDIR13, předpokládá se, že i fylogeneticky nejbližší protein AtDIR14 je potenciálně regulován cytokininy. Pro strukturní a in vitro funkční studie tohoto proteinu je nutné jeho dostatečné množství, a proto se v této bakalářské práci věnuji produkci funkční domény AtDIR14 proteinu. Byl připraven DNA konstrukt pro produkci funkční domény AtDIR14 v buňkách Pichia pastoris GS115. Metanolem indukovanou expresí rekombinantního proteinu v Erlenmeyerových baňkách a jeho purifikací z expresního média použitím ionexové a gelové filtrační chromatografie bylo získáno 360 µg l-1 AtDIR14.

Abstract:

Dirigent proteins in plants impart regio- and stereoselectivity to phenoxy radical coupling reactions and thus play an essential part in the synthesis of biologically active natural products, such as lignans and gossypol. Since expression of the AtDIR13 protein changes in response to cytokinin trans-zeatin and the ARR10 and ARR1 transcription factors, members of the cytokinin signaling pathway, have a high affinity for the AtDIR13 promoter, it is believed that even the phylogenetically closest AtDIR14 protein is possibly regulated by cytokinins. For structural and in vitro functional studies, enough protein is needed and therefore in this bachelor thesis I focus on the production of the functional domain of AtDIR14 protein. A DNA construct to produce the functional domain of AtDIR14 in Pichia pastoris GS115 cells was prepared. Methanol induced expression of the recombinant protein in Erlenmeyer flasks and its purification from expression media using ion exchange and gel filtration chromatography yielded 360 µg l-1 of AtDIR14.
