Effect of tumour bacteriome on survival of patients with colorectal carcinoma

Abstract:

Colorectal cancer, the second most common cause of cancer-related death worldwide, is associated with a poor prognosis in the advanced stages of the disease. Therefore, it is necessary to constantly deal with the possibilities of new therapeutic and prognostic procedures and strategies. According to recent research, the bacteria inhabiting the large intestine called the gut microbiome, are associated with the risk of developing this oncological disease. Describing the presence and structure of the microbiome in colon and rectal cancer could thus contribute to determining the prognosis and subsequent more effective treatment of these patients. This bachelor's thesis deals with the association of the community of bacteria identified with the help of 16SrRNA sequencing from tumour tissue smears with the overall survival of patients. The work focuses on the analysis of the composition of the colon and rectal tumour bacteriome of 202 patients. The results suggest that, in addition to the clinical stage of the disease, the composition of the bacterial community may be a potential biomarker for predicting the prognosis of patients with colorectal cancer. It points to specific bacterial families and genera that are associated with the microbiome of the healthy population in the available literature and could play a significant role in the context of colorectal cancer.

Abstract:

Kolorektální karcinom, jako celosvětově druhá nejčastější příčina úmrtí týkající se nádorových onemocnění, je ve vyšších stadiích onemocnění spojen se špatnou prognózou. Proto je nutné se neustále zabývat možnostmi nových terapeutických a prognostických postupů a strategií. Bakterie osidlující tlusté střevo, nazývané střevní mikrobiom, jsou dle recentního výzkumu spojovány s rizikem vzniku a vývojem tohoto onkologického onemocnění. Popsání přítomnosti a struktury mikrobiomu v souvislosti s karcinomem tlustého střeva a konečníku by tak mohlo přispět k určení prognózy a následné efektivnější léčbě těchto pacientů. Tato bakalářská práce se zabývá asociací společenstva bakterií identifikovaných s pomocí 16SrRNA sekvenování ze stěrů nádorové tkáně s celkovým přežitím pacientů. Práce se zaměřuje na analýzu složení bakteriomu nádoru tlustého střeva a konečníku 202 pacientů. Výsledky naznačují, že kromě klinického stadia onemocnění, může být složení bakteriální komunity potenciálním biomarkerem pro predikci prognózy pacientů s kolorektálním karcinomem. Poukazuje na konkrétní bakteriální čeledi a rody, které jsou v dostupné literatuře spojovány s mikrobiomem zdravé populace a v kontextu kolorektálního karcinomu by mohly hrát nezanedbatelnou roli.
