Molecular phylogeny of selected taxa of Sciaroidea (Diptera: Bibionomorpha), with emphasis on the family Keroplatidae

Abstract:

This dissertation thesis deals with molecular phylogeny of selected groups of the superfamily Sciaroidea (Diptera: Bibionomorpha). This superfamily includes nine recent families, which are Bolitophilidae, Cecidomyiidae, Diadocidiidae, Ditomyiidae, Keroplatidae, Lygistorrhinidae, Mycetophilidae, Rangomaramidae and Sciaridae. However, we must also include here species from the so called Sciaroidea incertae sedis, whose family assignment is not clear yet. The larvae of Sciaroidea are mostly mycophagous, feeding either on fungal mycelia in dead wood or inside the fruit bodies of various fungi, with several groups being predators of other arthropods, or phytophagous, forming galls on higher plants, as is the case of the family Cecidomyiidae. The main aim of this thesis is to reconstruct phylogeny of various subgroups of Sciaroidea. Various molecular markers, either mitochondrial (12S, 16S, COI, cytb) or nuclear (18S, 28S, CAD, MCS), were used. The results of particular studies mostly correspond to previously established relationships based on morphological characters, but we have also found new and unexpected phylogenetic relationships, which are discussed here. This dissertation is also focused on the taxonomy of the family Keroplatidae. Altogether 4 new species and 3 new genera were described in the papers included in this thesis.

Abstract:

Předložená dizertační práce se věnuje molekulární fylogenezi vybraných skupin nadčeledi Sciaroidea (Diptera: Bibionomorpha). Tato nadčeleď zahrnuje devět recentních čeledí, kterými jsou, Bolitophilidae, Cecidomyiidae, Diadocidiidae, Ditomyiidae Keroplatidae, Lygistorrhinidae, Mycetophilidae, Rangomaramidae a Sciaridae. Nicméně zde také musíme zařadit druhy z tzv. skupiny Sciaroidea incertae sedis, jejichž zařazení do čeledi není dosud zcela jasné. Larvy z nadčeledi Sciaroidea jsou většinou mykofágní, živící se buď houbovými mycéliemi v odumřelých stromech či plodnicemi různých druhů hub, dále se zde můžeme setkat s predátory jiných členovců nebo se larvy mohou živit fytofágně, kdy tvoří hálky na tělech vyšších rostlin, jako je tomu v případě čeledi Cecidomyiidae. Hlavním cílem této práce je rekonstruovat fylogenezi různých podskupin nadčeledi Sciaroidea. Bylo použito různé množství genetických markerů, ať už těch mitochondriálních (12S, 16S, COI, cytb) nebo také jaderných (18S, 28S, CAD, MCS). Výsledky dílčích studií povětšinou korespondují s již dříve stanovenými vztahy založených na morfologických studiích, ovšem objevili jsme i nové a nečekané vztahy, které jsou v této prací diskutovány. Zároveň je tato práce zaměřena na taxonomií čeledi Keroplatidae. Celkem byly popsány 4 nové druhy a 3 nové rody v publikacích zahrnutých v této práci.
