Neuroinflammation in the trigeminal system

Abstract:

Trigeminální neuropatická bolest způsobená poškozením nervus trigeminus je v porovnání s ostatními typy neuropatické bolesti velmi intenzivní a extrémně zatěžující. Zvířecí modely jsou zásadní pro pochopení mechanismu neuropatické bolesti a pro vývoj účinné terapie. Většina dosud publikovaných experimentálních výsledků popisujících buněčné a molekulární změny vedoucí k rozvoji neuropatické bolesti je založena na modelu poranění míšních nervů. Dosavadní experimentální studie zabývající se mechanismy vzniku trigeminální neuropatické bolesti jsou velmi omezené. Experimentální modely trigeminální neuropatické bolesti jsou založeny na poškození jedné z větví nervus trigeminus. Tento zásah vyvolává buněčné a molekulární změny v trigeminálním gangliu (TG), které jsou spojeny s rozvojem neurozánětu. Mimo to mohou podobné změny probíhat při synapsích a v okolí druhých neuronů trigeminální dráhy, které jsou uloženy v nucleus spinalis n. trigemini nacházející se v prodloužené míše. Cílem předložené dizertační práce je studium buněčných změn a změn v expresi chemokinů CCL2 a CX3CL1 (fraktalkinu) a jejich receptorů CCR2 a CX3CR1 v trigeminálním gangliu a v nucleus spinalis n. trigemini po unilaterální ligatuře nervus infraorbitalis.

Abstract:

Trigeminal neuropathic pain caused by damage to the trigeminal nerve is very intense and extremely stressful compared to other types of neuropathic pain. Animal models are essential for understanding the mechanism of neuropathic pain and for developing effective therapy. Most of the published experimental results describing cellular and molecular changes leading to the development of neuropathic pain are based on a model of spinal nerve injury. Previous experimental studies dealing with the mechanisms of trigeminal neuropathic pain are very limited. Experimental models of trigeminal neuropathic pain are based on damage to one of the branches of the trigeminal nerve. This intervention induces cellular and molecular changes in the trigeminal ganglion (TG), which are associated with the development of neuroinflammation. In addition, similar changes may occur at synapses and around other neurons of the trigeminal pathway, which are located in the nucleus spinalis of the trigeminal nerve located in the medulla oblongata. The aim of the presented thesis is to study cellular changes and changes in the expression of chemokines CCL2 and CX3CL1 (fractalkine) and their receptors CCR2 and CX3CR1 in the trigeminal ganglion and in the trigeminal subnucleus caudalis after unilateral ligature of the infraorbital nerve.
