Metagenomics in the study of resistance of bacteria to antimicrobial agents

Abstract:

Mikroorganismy rezistentní vůči antimikrobiálním látkám představují globální zdravotnický problém. Většina mikroorganismů nemůže být kultivována v laboratorních podmínkách, a tak rezervoár genů rezistence v nekultivovatelných mikroorganismech zůstává poměrně neprozkoumán. Pro studium nekultivovatelných mikroorganismů byl vyvinut nový vědní obor metagenomika. Bakalářská práce je zaměřena na využití metagenomiky při studiu genů rezistence ke klinicky významným skupinám antibiotik v různých prostředích. Součástí práce je stručný popis mechanismů rezistence a metodických přístupů včetně sekvenačních platforem, které jsou nedílnou součástí metagenomové analýzy. Poslední část bakalářské práce je věnovaná aktuálním poznatkům z výzkumu genů rezistence v mikrobiomu člověka, zvířat, vody, půdy a vnějšího prostředí. Z výsledků studií vyplývá, že výskyt a diverzitu genů rezistence ovlivňuje zejména selekční tlak antimikrobiálních látek.

Abstract:

Microorganisms resistant to antimicrobial agents represent a global health concern. The vast majority of microorganisms cannot be cultivated under laboratory conditions, so the reservoir of resistance genes in uncultured microorganisms is still relatively unexplored. For research of uncultivated microorganisms was developed a new scientific field – metagenomics. This bachelor’s thesis is focused on use of metagenomics in the study of resistance genes to clinically important antibiotics in different environments. The thesis also includes brief overview of resistance mechanisms and methodical approaches including sequencing platforms which are an integral part of metagenomic analysis. The last part of the bachelor’s thesis contains current findings from research on resistance genes present in human, animal, water, soil and environmental microbiomes. The results show that the abundance and diversity of resistance genes are affected mainly by selection pressure of antimicrobial agents.
