The use of retinoids in differentiation therapy of human malignancies

Abstract:

Deriváty vitaminu A (retinolu), retinoidy, mají nezastupitelnou úlohu ve vývoji organismu. Jsou to důležité mediátory růstu a diferenciace, uplatňují se v procesech embryogeneze a morfogeneze. Retinoidy regulují vývoj a růst normálních i maligních buněk prostřednictvím jejich vazby na jaderné receptory, které následně fungují jako inducibilní transkripční faktory. Pro svoji schopnost indukovat buněčnou diferenciaci jsou tyto deriváty retinolu již řadu let využívány v medicíně, zejména v dermatologii a v prevenci i léčbě nádorových onemocnění. Principy působení retinoidů na maligné buňky spočívají v indukci diferenciace, v inhibici proliferace nebo indukci apoptózy. Jejich zavedení do klinické praxe představuje výrazný pokrok v léčbě malignit. Cílem bakalářské práce je poskytnout základní přehled možností léčebného využití retinoidů v různých oborech medicíny a shrnout aktuální poznatky o používání retinoidů v rámci diferenciační terapie.

Abstract:

Derivatives of vitamin A (retinol), retinoids, have an unsubstitutable function in organism development. Retinoids are important mediators of cellular growth and differentiation, assert their influence in processes of embryogenesis and morphogenesis. Retinoids modulate the growth of normal and malignant cells through their binding to retinoid nuclear receptors and their subsequent activation. Because of their ability to induce cell differentiation, these derivatives of retinol are used in medicine, especially in dermatology and in prevention or therapy of cancer. Retinoids effect on malignant cells through the differentiation induction, proliferation inhibition or apoptosis induction. Their usage in clinical praxis constitutes a distinctive progress cancer therapy. This bachelor thesis reviews retinoids therapy opportunities in different fields of medicine and gives a summary of actual findings about the using of retinoids in differetiation therapy.
