Molecular detection of zoonotic microorganisms in free living rodents

Abstract:

Hlodavci jsou nejpočetnějším řádem savců na světě, který obsahuje na 2000 druhů. Mnohé z nich jsou také významnými hostiteli mnoha patogenů. Udává se, že se podílejí na přenosu více než 85 zoonotických patogenů, a to nejen virů a bakterií, ale také hub, protozoí či helmintů. Cílem diplomové práce bylo stanovit prevalenci vybraných patogenů (Leptospira sp., Bartonella sp., Rickettsia sp., Borrelia miyamotoi) pomocí molekulárních metod v populaci hlodavců odchycených v okolí Valtic v létě a na podzim 2014. Celkem bylo zpracováno 133 vzorků ledvin a 127 vzorků slezin ze 136 hlodavců druhů hraboš polní (Microtus arvalis), norník rudý (Myodes glareolus), a myšice (myšice křovinná - Apodemus sylvaticus, myšice lesní - Apodemus flavicollis). V této práci se podařilo zachytit DNA pouze 2 z těchto agens a to leptospir (15,4 %) a bartonel (15,4 %). Tato práce doplňuje chybějící informace o těchto patogenech u hlodavčích hostitelů v České republice a může sloužit jako základ pro obdobné navazující výzkumy. Prevalenční studie u savčích hostitelů i artropodních vektorů přispívají ke sledování výskytu daného onemocnění a k porozumění vzájemných vztahů mezi populací savčího rezervoáru, vektoru a počtem případů daného onemocnění u lidí.

Abstract:

Rodents are the most numerous order of mammals worldwide. It consists of about 2000 species. Some of them serve as an important host of many pathogens. They participate in the transmission of more than 85 zoonotic pathogens - viruses, bacteria, fungi, protozoa or helminths. The goal of this thesis was to determinate the prevalence of chosen pathogens (Leptospira sp., Bartonella sp., Rickettsia sp., Borrelia miyamotoi) with molecular methods (PCR) in the population of rodents caught near Valtice town in summer and autumn 2014. In total 133 kidney and 127 spleen samples from 136 animals of 4 species (Microtus arvalis, Myodes glareolus, Apodemus sylvaticus, Apodemus flavicollis) were tested. We detected DNA of only two pathogens – Leptospira spp. (15,4 %) and Bartonella sp. (15,4 %). This thesis completes missing information about these pathogens in rodent hosts in the Czech Republic. It may serve as the basis for similar following studies. The screening studies on mammal hosts and arthropod vectors may help to discover the distribution of the pathogens, relations between a population of mammal reservoir, arthropod vector and the number of cases of the disease in humans.
