Expression and activity of MDR-associated ABC transporters in mES cells

Abstract:

Mnohočetná léková rezistence (multidrug resistance - MDR) je způsobena nadměrnou expresí ABC transportérů, které chrání kmenové buňky před toxickými látkami ať už endogenního nebo exogenního původu. Předpokládá se, že kmenové buňky osidlují prostředí s nízkou koncentrací kyslíku. Proto jsme se rozhodli testovat, jak je hypoxie zapojena do regulace MDR transportéru a současně, jaký význam MDR transportéry pro kmenové buňky mají. Vystavení embryonálních kmenových buněk hypoxii mělo u studovaných MDR transportérů za následek snížení jejich exprese, jež se zdála závislá na stabilizaci transkripčního faktoru HIF1α. V dalších experimentech jsme zjistili, že inhibice MDR transportérů vedla k diferenciaci myších neurálních kmenových buněk a to jak primitivních, připravovaných diferenciací z mES buněk, tak i definitivních z mozků myších embryí. Je tedy jasné, že MDR transportéry nejsou jen pouhou ochranou kmenových buněk, ale také se účastní fyziologických dějů jako je diferenciace, maturace nebo proliferace a jejich aktivita je zřejmě důležitá v zachování kmenovosti jako takové.

Abstract:

Multidrug resistance (multidrug resistance - MDR) is caused by ABC transporters, which protect the stem cells from toxic substances of either endogenous or exogenous origin. It is believed that stem cells colonize the environment with a low oxygen concentration. That is why, we decided to test how hypoxia is involved in the regulation of MDR transporters and simultaneously, we tested the importance of MDR transporters for stem cells. Exposure of embryonic stem cells to hypoxia resulted in decreased expression of studied MDR transporters and their expression appeared to depend on the stabilization of transcription factor HIF1α. In further experiments, we decided to investigate if MDR transporters have any other effect in stem cell fate determination. We showed that inhibition of MDR transporters leads to differentiation of mouse neural stem cells, both primitive (prepared by differentiation of mES cells) and definitive, which were the final neural stem cells from brains of mouse embryos. It is clear that the MDR transporters do not only protect stem cells, they also participate in physiological processes such as differentiation, maturation and proliferation, and their activity is probably important in maintaining stemness as such.
