Abstract:

Práce se zabývá využitím separačních metod a blízké infračervené spektroskopie při hodnocení kvalitativních parametrů řepky olejné. Prvním cílem disertační práce byl vývoj minimalizační metody stanovení obsahu mastných kyselin. Byla vytvořena, optimalizována a validována metoda stanovení methylesterů mastných kyselin pomocí plynové chromatografie z minimálního množství výchozího vzorku (1 semeno, segment semene řepky olejné a dělohy mikrosporových embryí). Metoda byla aplikována do šlechtitelské praxe, v rámci které se analyzovaly obsahy mastných kyselin tzv. "high oleic" nebo "high oleic low linolenic" řepek. Možnost včasné selekce požadovaných genotypů povede k významnému zefektivnění výběru perspektivních šlechtitelských linií s požadovanou skladbou mastných kyselin a ohledem na kvalitu vyprodukovaného oleje. Další aplikací metody byly analýzy mastných kyselin v dělohách mikrosporových embryí při tvorbě dihaploidních regenerantů a v materiálech pocházejících z resyntézy řepky. Druhým cílem práce bylo stanovení obsahu glukosinolátů v semenech a zelené hmotě řepky olejné s použitím vysokoúčinné a ultra účinné kapalinové chromatografie. Původní metoda stanovení glukosinolátů pomocí kapalinové chromatografie podle normy ČSN EN ISO 9167-1 byla modifikována s cílem odstranění časové náročnosti. Byl proto vytvořen optimalizovaný postup stanovení glukosinolátů pomocí extrakce na pevné fázi a separace glukosinolátů ve formě desulfoglukosinolátů pomocí vysokoúčinné a ultra účinné kapalinové chromatografie. Vyvinutá metoda byla rovněž validována a aplikována při analýze obsahu glukosinolátů v zelené hmotě řepky. Účelem analýz zelené hmoty bylo identifikovat souvislost mezi poškozením rostlin řepky olejky třemi nejdůležitějšími škůdci (dřepčík, blýskáček a bejlomorka) a obsahem glukosinolátů v zelené hmotě. Třetím cílem práce bylo ověření uplatnění blízké infračervené spektroskopie pro analýzu řepky olejné v rámci šlechtění a hodnocení genových zdrojů kolekce olejnin Národního programu konzervace a využívání genetických zdrojů rostlin a agrobiodiverzity. Byly vyvinuty kalibrační modely pro analýzu rostlinného materiálu řepky olejky a otestován potenciál této techniky pro predikci obsahu oleje, sušiny, glukosinolátů a hlavních mastných kyselin ve vzorcích semen. Použití této metody je časově efektivní, nenáročné na obsluhu, vzorky není třeba nijak upravovat a lze změřit mnoho vzorků za krátký interval. Ve většině případů byly vyvinuty velmi dobré kalibrace, jejichž korelační koeficienty byly v rozmezí od 0,868 - 0,996. Některé kalibrace byly méně přesné, ale přesto použitelné pro orientační stanovení hodnot při šlechtění řepky. Vyvinutí robustních, přesných a spolehlivých kalibračních modelů umožnilo analýzu řepkových semen v krátkém časovém úseku ihned po sklizni. Pro následný zásev ještě v témže roce na základě analýz proběhla selekce perspektivních šlechtitelských materiálů. Neperspektivní materiály s nevyhovující kvalitou byly ze šlechtitelského procesu vyřazeny, což by vedlo k jeho značnému odlehčení a zefektivnění.

Abstract:

The thesis deals with the usage of separation methods and near infrared spectroscopy in evaluation of the qualitative parameters of rapeseed. The first goal of the dissertation was to develop a minimalistic method for the determination fatty acid content. The method for the determination the fatty acid methyl esters by gas chromatography from a minimum amount of the initial sample (1 seed, rape seed segment and microspore embryo cotyledons) was developed, optimized, and validated. The method was applied to breeding practices of analysing the content of fatty acids in "high oleic" nebo "high oleic low linolenic" rapeseed. The possibility of timely selection of desired genotypes will lead to a significant improvement in the choice of prospective breeding lines with the desired fatty acid composition and the quality of the produced oil. Another application was represented by the analysis of fatty acids in the microspore embryo cotyledons during forming dihaploid regenerants and in the materials from rapeseed resynthesis. The second aim of the work was determination the content of glucosinolates in the seeds and the green matter of rapeseed oil using high-perfomance and ultra-perfomance liquid chromatography. The original method for the determination glucosinolates by liquid chromatography according to the standard ČSN EN ISO 9167-1 was modified with the aim of reducing the time required. Therefore, the optimized procedure for the determination glucosinolates by solid phase extraction and separation of glucosinolates in a form of desulfoglucosinolates by high-perfomance and ultra-perfomance liquid chromatography was developed. The developed method was also validated and applied in the analysis of the glucosinolate content in the green matter of rapeseed. The purpose of the green matter analysis was to identify the relation between the damage of the rapeseed plants by the three most important pests (epitrix, meligethes and gall midge) and the content of glucosinolates in the green matter. The third aim of the thesis was to verify the application of near infrared spectroscopy for the analysis of rapeseed in the process of breeding, and to evaluate the collection of oilseed crops of The Czech National Programme on Conservation and Utilization of Plant Genetic Resources and Agro-biodiversity. The calibration models were developed to analyse the rapeseed plant material and the potential of this technique to predict the content of oil, dry matter, glucosinolates and major fatty acids in the seed samples was tested. The usage of this method is time-efficient, easy to handle, there is no need to modify samples; in addition, many samples can be analyzed in a short interval. In the most cases, very good calibrations with the correlation coefficients ranging from 0.868 to 0.996 were developed. Althoug some calibrations exhibited a lower accuracy, they were still useful for a rough evaluation in the process of breeding. The development of the robust, accurate, and reliable calibration models has enabled rapeseed to be analysed in a short time period immediately after harvesting. On the basis of the analyses, the selection of the promising breeding material was carried out for the subsequent cultivation in the same year. The materials of unsatisfactory quality were eliminated from the breeding process, which should lead to its higher efficiency.
