Epigenetic mechanisms in the regulation of telomere maintenance

Abstract:

Telomery jsou nukleoproteinové struktury, které se nacházejí na koncích lineárních eukaryontních chromosomů. Slouží jako jejich ochrana před rozpoznáním reparačními mechanismy buňky, před fúzemi chromozomů a tím i před dalšími genomovými nestabilitami. Jelikož replikační aparát opožďujícího se řetězce skrze Okazakiho fragmenty nemůže plně syntetizovat 3’ konec DNA, je přirozenou vlastností telomer se během života zkracovat. V buňkách, které se účastní časté replikace, je za doplňování konců chromozomů zodpovědný enzymatický komplex telomerázy. Strukturní a funkční regulace telomer je nezbytně nutná pro správné fungování jednotlivých buněk i celého organismu, neboť nefunkční, kriticky zkrácené telomery mohou vést až k nenávratnému zastavení buněčného cyklu, apoptóze, a tím i k předčasnému stárnutí organismu. Naopak stabilně udržované telomery jsou jednou ze základních podmínek buněčné nesmrtelnosti a možného rozvoje nádorového bujení. Studium epigenetické regulace telomer se rozšířilo v průběhu první dekády tohoto století a přineslo některé protichůdné teze, které zde budou diskutované. Tato práce se tedy zabývá osvětlením některých nekonzistentních výsledků za použití méně tradičních rostlinných modelových organismů jako například Nicotiana tabacum (tabák viržinský) nebo Ballantinia antipoda. Je zde popsáno udržování telomer v rostlinách s poškozenou methylací histonů a/nebo DNA, dále se práce zabývá i popisem epigenetického prostředí pravých – koncových – telomer a interních telomerových repetic. Na samém závěru jsou diskutovány některé hypotézy udržování telomer v rostlinách, které představuje sama autorka.

Abstract:

Telomeres are nucleoprotein structures located at the ends of linear eukaryotic chromosomes. They prevent natural chromosome ends from unwanted DNA damage response, fusions and consequent onset of genome instability. Since the replication of lagging strand via Okazaki fragments cannot fully replicate 3’ end of chromosome, the natural attribute of telomeres is their shortening in each cell cycle during the life of organism. In cells that undergo frequent divisions, the lengthening of telomeres is provided by enzymatic ribonucleoprotein complex called telomerase. Structural and functional regulation of telomeres is essential for correct cell and whole organism development – non-functional, critically shortened telomeres may stop the cell cycle and lead to apoptosis and premature ageing of organism, while stably maintained telomeres are one of the crucial conditions for cellular immortality and possible cancer development. The study of epigenetic regulation of telomeres became popular during past decade resulting in contradictory findings that are summarized in this work. We thus decided to shed light on some of the inconsistent data using less common plant model organisms such as Nicotiana tabacum or Ballantinia antipoda. In this thesis, I describe telomere maintenance in plants with disrupted histone and/or DNA methylation, and epigenetic landscape of genuine – terminal – telomeres and internal telomeric repeats (ITR). In the end, we discuss possible regulatory pathways in the maintenance of plant telomeres via epigenetic mechanisms
